Cells were lysed in Radioimmunoprecipitation (RIPA) buffer supplemented with protease inhibitors (Life Technologies, 87786) and centrifuged. Protein concentration in the supernatant was measured using a Bicinchoninic acid assay (BCA) protein assay kit (Bio-Rad). Proteins were resolved on 4 to 12% NuPAGE Bis-Tris gels (Fisher Scientific) and transferred to Polyvinylidene difluoride (PVDF) membranes (Bio-Rad). Western blotting was performed using a Pierce Fast Western Blot Kit (Thermo Scientific) according to the manufacturer’s protocol with the following primary antibodies: GAPDH (Proteintech, HRP-60004), PCIF1 (Proteintech, 16082-1), ACE2 (Novus, NBP2-67692), TMPRSS2 (Abcam, ab92323), and NRP1 (Abcam, ab81321).

ACE2 and Syndecan-1 Expression in Cells

HEK293T hACE2 and VeroE6 cells were treated with 30 μM KSL-128114 or DMSO for 6 h, and then fixed in 4% formaldehyde. Cells were then either permeabilized in 0.5% Triton X-100 or left unpermeabilized. Then, cellular expression of ACE2 or Syndecan-1 were detected by rabbit monoclonal antibodies (ACE2, Novus bio, NBP2-67692, 1:500, Syndecan-1, abcam, ab128936 2 μg/mL) and secondary donkey anti-rabbit Alexa Fluor 488 (1:2000, Thermo Fisher Scientific). Cellular fluorescence was measured using a TROPHOS plate RUNNER HD.
