Dulbecco’s Modified Eagle’s Medium (DMEM), Ham’s nutrient mixture F-12, and fetal bovine serum (FBS) were purchased from Life Technologies (Gibco BRL, Grand Island, NY, USA). Palmitate and 4-(2-aminoethyl)benzenesulfonyl fluoride hydrochloride (AEBSF) were obtained from Sigma-Aldrich (St. Louis, MO, USA). The Palmitate was prepared as described previously [30 (link)]. ASYM24 antibody (#07-414) was obtained from Millipore. β-actin antibody (sc-1616) and p-PERK antibody (sc-32577-R) were purchased from Santa Cruz Biotechnology (Santa Cruz, CA, USA). All reagents were of the highest purity commercially available.

GFP-ALY Protein Purification from 293T Cells

made a stable cell
line from 293T cells expressing GFP-ALY growing in DMEM medium plus
10% fetal bovine serum. With this cell line, we made whole cell extract
with H lysis buffer (150 mM NaCl, 20 mM Tris, pH 7.8, 0.5 mM ETDA,
0.2 mM PMSF, 1 mM NaF, 1 mM sodium orthovanadate, 1% Np-40, 1 mM DTT).
The cells in H lysis buffer were incubated on ice for 30 min before
being cleaned by spinning at 12000g for 20 min. Conjugated
GFP antibody beads (Allele Biotech Inc., San Diego, CA) were used
to pull down GFP fusion protein. The samples were dissolved in SDS–PAGE
gels and detected by ASYM24 antibody (Millipore) in Western blot assay
(chemiluminescence reagents).
