Vessel Density Quantification in Tumor Sections

Tumor sections (5 μm), two sections per tumor, were stained with anti-PECAM-1 (M-20) (Santa Cruz Biotechnology). Bound antibody was detected with ImmPRESS (Peroxidase) Polymer anti-rabbit immunoglobulin reagent (Vector Laboratories) and visualized using ImmPACT DAB peroxidase (HRP) substrate (Vector Labs). Mayer's hematoxylin (Sigma-Aldrich) was used as a counterstain. Slides were digitized using an Aperio ScanScope XT scanner (Leica), and computer-aided image analysis was performed and manually checked for quality assurance. Regions of interest were identified with an algorithm that distinguishes tumor from stroma and (peri-)necrotic regions. The vessel density within the tumor region, as well as in the adjacent normal tissue, was quantified using the Definiens Tissue Studio software platform (Definiens). Quantification was done while blinded to the treatment groups.

Quantification of Murine Pancreatic Islets

Freshly dissected murine pancreas tissues were fixed in 4% paraformaldehyde (P6148; Sigma-Aldrich) for 16 h at 4°C, embedded with paraffin and stained with H&E. H&E-stained tissue slides were scanned using Aperio ScanScope XT scanner (Leica). Islet number on each pancreas section was quantified by ImageScope software (Leica v12.3.3.5048). A total of 24 pancreas sections (four sections/mouse with at least 50 μm apart from six mice/group) were included in the quantification of islet numbers.

Quantitative Analysis of Ki67 Expression

Expression of Ki67, the protein product of the Mki67 gene was validated using immunohistochemistry with antigen retrieval (1X Citra Plus Buffer; Biogenex, Fremont CA; 30 min in a steamer), monoclonal rat anti-mouse Ki67 (TEC3; 1:50; 60 min; Dako, Carpinteria, CA) followed by biotinylated rabbit anti-mouse IgG (1:200; 30 min; Dako). Staining was visualized with streptavidin/HRP (1:200; 30 min) and DAB (10 min; Dako) and counterstained with Mayer's hematoxylin (2 min; Sigma-Aldrich). Staining was quantified from scanned images (Aperio Scanscope XT scanner, Leica, Buffalo Grove, IL) using positive pixel counting [ImageScope 8.1 (Aperio®) digital image analysis package]. The Ki67 protein expression is expressed as a percentage of positive pixels relative to the total (positive + negative) pixels for a whole AP cross-section.
