Trypan blue, Alamar blue, Sulforodamine B and cell culture mediums were purchased from Sigma-Aldrich (St. Louis, MO, USA). The Smart Button Data loggers were purchased from ACR System Inc (Surrey, Canada).

Evaluating Anti-Inflammatory and Estrogenic Effects

Adipose Stem Cell Viability Assays

Adipose derived stem cells were plated at density of 3 × 103 per well in 96-well plates and incubated for 72 h. After that WST-1 (Roche, Switzerland) and sulforodamine B (SRB, Sigma-Aldrich, USA) assays were done.
For WST-1 absorbance of dye was measured at a wavelength of 450 nm and for SRB assay absorbance of dye was measured at a wavelength of 590 nm in 96-well plate (Thermo Fisher Scientific, USA) using Microplate Reader Perkin Elmer Wallac Victor 2 (Perkin Elmer, USA). All the tests were performed in septuplicate.

SRB Viability Assay for Cell Cytotoxicity

Sulforodamine B, trichloroacetic acid and acetic acid were purchased from Sigma Aldrich; Merck KGaA. A total of 3x103 cells were plated per/well in a 96-well plate and incubated overnight. Subsequently, cells were treated in triplicates as described above and cultured for 48 h. A SRB viability test was performed as follows: Cells were fixed for 1 h on ice with 10% trichloroacetic acid (v/v in RPMI-1640), washed trice with double distilled water, and dried and stained for 1 h at room temperature with 0.057% Sulforodamine B (w/v in 1% acetic acid). After staining, the plates were washed three times with 1% acetic acid and then dried, and 200 µl 10 mM Tris was added to each well to solubilize the SRB. The absorbance was measured at 570 nm using an ELISA reader (Biotek Instruments, Inc.). Each experiment was repeated at least three times.

Sulforhodamine B Cytotoxicity Assay

24, 48 and 72 h after HCP addition the culture medium was discarded and cells were fixed with 50% trichloroacetic acid (Sigma Aldrich, Poland,; 100 μl per well, 1 h, 4 °C). Fixed cells were washed 3 times in water, air dried and 100 µl of sulforodamine B (0.4% in 1% acetic acid; Sigma Aldrich, Poland) was added. After 30 min incubation cells were washed with 1% acetic acid 4 times, air dried and 10 mM Trisma®base (100 µl/well; Sigma Aldrich, Poland) was added to dissolve the pigment. The fluorescence was measured at excitation 570 nm (± 10) and emission 590 nm (± 10) using FLUOstar Omega reader (BMG Labtech, Sweden).
