Sciatic Nerve Atrophy Evaluation

In order to assess the degree of atrophy of the muscles innervated by the operated left sciatic nerve, the weight (w) and volume (v) of the whole lower leg was measured at 14 weeks after surgery. Lower legs were harvested after the intracardiac perfusion (see details below). The lower legs [which contain several muscles exclusively innervated by the sciatic nerve trunk (Greene, 1968 )] were exposed and then disjointed from the knee and ankle. For the w assessment, the dissected legs were removed from the fixative, dried in absorbent paper and weighed in a digital weighing machine (Sartorius BP 121S, precision: 0.1 mg, Sigma–Aldrich). For the v assessment, dissected legs were immersed in test tube (50 ml) containing 30 ml of PBS and the increase of this volume represented the lower legs volume. In this study, the percentage of w and v loss was calculated between the operated and the contralateral-side leg from each animal, including healthy animals (CTR-Native), and these values were used for statistical comparisons.

Muscle Atrophy Quantification Protocol

To assess the degree of muscle atrophy, the whole lower leg was separated from the surrounding skin and dissected from the knee and ankle then fixed immediately in 10% buffered formalin for at least 1 week. The legs were weighted using a digital balance (Sartorius BP121 S, precision level = 0.1 mg, Sigma-Aldrich). The volume was measured following Archimede’s principle and fluid displacement rules, where a cylinder was filled with 30 mL PBS and the leg was immersed totally in the cylinder, the volume of the immersed leg is equal to the volume of liquid displacement. For each rat, the operated leg and contralateral leg were measured, and the percentage of loss was calculated as [100 – (operated leg/contralateral leg × 100)].

Thermal Analysis of HC Samples

The TGA study was carried out on a NETZSCH STA 449 F1 Jupiter simultaneous thermal analysis instrument (Selb, Germany). The HC samples were analyzed in argon at a heating rate of 10 °C min−1, temperatures from 30 to 900 °C, and protective and blowout gas flow rates of 20 and 50 mL min−1, respectively. The Al2O3 cylindrical crucible with a perforated cover was used and a reference was an empty corundum crucible with a cover.
