Quantifying cDNA Concentration and Size

Check if the same lab product or an alternative is used in the 5 most similar protocols
Sample cDNA concentration was measured using the Quant-iT PicoGreen dsDNA assay kit and black Nunc F96 MicroWell polystyrene plates (ThermoFisher Scientific) according to the manufacturer’s instructions, except that we used half of the recommended reaction volumes to reduce costs. Fluorescence measurements were recorded on a Hidex Sense instrument (Hidex). To assess cDNA size distribution, eleven samples were randomly selected from each plate, loaded on a HS DNA chip and run on a 2100 Bioanalyzer System (software vB.02.10.51764, Agilent).

cDNA Size Distribution Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Sample concentrations were quantified by using the Quant-iT™ PicoGreen™ dsDNA Assay Kit (ThermoFisher Scientific) and performed in black Nunc™ F96 MicroWell™ Polystyrene Plates (ThermoFisher Scientific), according to the manufacturer's instructions, except that all reaction volumes were cut by half to reduce costs. Fluorescence measurements were recorded on a Hidex Sense instrument (Hidex). In order to assess cDNA size distribution, 11 samples from each plate were randomly chosen, loaded on a High Sensitivity DNA chip and run on a 2100
Bioanalyzer System (Agilent Technologies). As a rule of thumb, the criteria for deciding whether an experiment was worth to be followed up were the following: average size distribution around 1.5-2.0 Kb, low proportion of short fragments (<400 bp) and absence of residual primers/primer dimers (<200 bp).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
