After Cuc B incubation, cells and transfected cells were washed with PBS and fixed with 70% ethanol solution overnight at −20°C. After PBS washes the cells were stained with 5 µg/ml RNase and 20 µg/ml PI (propidium iodide/RNase A staining buffer, Invitrogen) in the dark at 37°C for 30 min and then analyzed with a flow cytometry (Becton Dickinson FACS Canto™, Franklin Lakes, NJ). At least 10,000 events were counted for each sample. The DNA content in the G0/G1, S, and G2/M phases were analyzed using ModFit 161 LT version 3.0 software (Verity Software House, Topsham, USA).

Cell Cycle Analysis by Flow Cytometry

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cells seeded into 6-well plates were incubated with indicated concentrations of LCA for 24 h. Cells were harvested, washed with PBS, as well as fixed in 70% ethanol and stored at 4 °C overnight. Cells were then stained in PBS containing 5 μg/mL RNase and 20 μg/mL PI in the dark at room temperature for 30 min and analyzed using a flow cytometry (Becton Dickinson FACS Canto, Franklin Lakes, NJ). 10,000 events were recorded for each sample. The DNA content was analyzed using ModFit 161 LT version 3.0 software (Verity Software House, Topsham, USA).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
