RBC Glutathione Quantification by LC-MS

RBC-GSH concentration was measured by an exogenous isotope dilution method using [Gly-13C2, 15N]-GSH (Cambridge Isotope Laboratories) as an internal standard. Briefly, 100 μL of the baseline RBC-free sample was spiked with a known quantity of isotopically labeled [Gly-13C2, 15N]-GSH. The samples were then analyzed by liquid chromatography mass spectrometry as described above. The spiked RBC-GSH was analyzed by selected reaction monitoring at precursor ions m/z 308 and 311 to product ions m/z 162 and 165, respectively.

Isotopically labelled molecules Cys* ( 13 C3 99%, 15 N 99%), NAC* ( 13 C3 97-99% 15 N 97-99%), GSH* ( 13 C2 98% 15 N 96-99%) and dGuo* ( 15 N5 98%) were purchased from Cambridge Isotope Laboratories (Andover, USA).
