Enantioselective Enzymatic Resolution of Racemic Ketoprofen

Racemic ketoprofen was isolated from inexpensive, over-the-counter tablets (100 mg PiSA Farmaceutica). (S)-(+)-ketoprofen analytical standard, Cándida rugosa lipase, Cándida antarctica lipase type B, porcine pancreatic Lipase, and Mucor javanicus lipase were obtained from Sigma Aldrich Company (St. Louis, MO, USA). Decan-1-ol, octan-1-ol, butan-1-ol, and methan-1-ol were purchased from Alfa Aesar. Cyclohexane, hexane, hexanes, isopropanol, and isooctane were purchased from Tedia (Ohio, OH, USA). Monobasic potassium phosphate and dibasic potassium phosphate were purchased from Spectrum Chemical Company. All other chemicals and analytical-grade reagents were from commercial sources and used as received.

Analytical Standards for Lipid Analysis

The following analytical standards were acquired from Cambridge Isotope Laboratories (5MChr), ChemService (DaiP), Fluka (BjF, DalP, DaeP, DahP), Supelco Inc. (BaA, Chr, BbF, BkF, BaP, DahA, IcdP and 37 component FAME mix) and Sigma (5α-cholestan, cholesta-3,5-diene, campesterol, stigmasterol, β-sitosterol, brassicasterol). High-performance liquid chromatography (HPLC)-grade hexane, cyclohexane, isopropanol and N,N-dimetilformamide were from Tedia, acetonitrile from J.T. Baker, methanol from Mallinckrodt and water was obtained from a Millipore Milli-Q purification system. Millex HV filters (0.45 µm) were purchased from Millipore and solid-phase extraction (SPE) columns were from Waters (Sep Pak C18, 500 mg, 3 mL).

Extraction and Quantification of PAHs

Hexane, cycloHexane, acetone, and dichloromethane (all HPLC-spectro grade) were purchased from Tedia (TC, USA). Internal standard hexamethylbenzene (99% purity) was acquired from Dr•Ehrenstorfer (Augsburg, Germany). The surrogate standard mix was acquired from AccuStandard (CT, USA), and contained naphthalene-d10, acenaphthene-d10, phenanthrene-d10, chrysene-d12 and perylene-d12 (4 mg ml -1 ). A standard solution of 16 US EPA PAHs, and the non-ionic surfactant Brij 30 [CH3(CH2)10CH2(OCH2CH2)nOH, were obtained from Sigma-Aldrich (St. Louis, MO).
