Inflammatory and Oxidative Markers in Kidney Tissue and Cells

TNF‐α (H052), IL‐1β (H002), IL‐6 (H007), and IL‐18 (H015) levels from mice kidney tissue and HK‐2 cells were measured using inflammation ELISA Kits (Nanjing Jiancheng Bioengineering Research Institute Co., Ltd, Nanjing, China). MDA (A003‐1‐2), SOD (A001‐3‐2), GSH (A006‐2‐1), and GSH‐PX (A005‐1‐2) levels from mice kidney tissue and HK‐2 cells were measured using MDA, SOD, GSH, and GSH‐PX kits according to the methods of manufacturer instructions (Nanjing Jiancheng Bioengineering Research Institute Co., Ltd, Nanjing, China). Absorbance was measured using UniCel DxC 600 Chemistry Analyzer (Beckman Coulter, CA, USA). ROS production from HK‐2 cells was assessed using an Olympus FluoView1000 confocal laser scanning microscope (using Ex/Em λ = 480 nm/535 nm).

Metabolic Profiling of Mouse Urine and Plasma

Mice were placed in a 24-hours metabolic cage for urine collection and urinary albumin levels were measured by a mouse albumin ELISA kit (Bethyl Laboratories, Montgomery, TX, USA). Urinary creatinine levels were assayed by a Creatinine Assay Kit (DICT-500; BioAssay Systems, CA, USA). The ratio of urinary albumin to creatinine was calculated to correct the difference in urine volume and expressed in micrograms per milligram. Before being killed, blood of mice was collected through retro-orbital vein under avertin anesthesia. Plasma cholesterol and triglyceride levels were determined using a UniCel DxC 600 Chemistry Analyzer (Beckman Coulter, CA, USA).

Renal Function Evaluation in Mice

Body weight and renal weight measurements of mice were accurate to 0.01 g with electronic scales. Kidney-to-body weight ratio was calculated as left or right kidney weight (g) divided by the body weight (kg). Biomarkers of Inflammation and Tissue Damage

Laboratory values (concentration of leukocytes, C-reactive protein (CRP), lactate dehydrogenase (LDH), creatine phosphokinase (CPK), and aspartate aminotransferase (AST)) were regarded as surrogate markers for inflammation and tissue destruction. The values of blood samples obtained from the day of bacterial sampling were observed. Serum Isolation and Liver Enzymes Analysis
