Comparative Hematological Analysis in Mice

Blood from 30- and 90-week-old RPE and wild-type mice (n = 3/group) was collected from the vein tail into lithium-heparin Microvette® 200 (Sarstedt). Samples were processed with the Sysmex XP-300™ Automated Hematology Analyzer (Sysmex Corporation, Japan) or treated with Red Blood Cell lysis solution (Qiagen). Thereafter, white blood cells were manually counted using a haemocytometer.

Biochemical Profile of Venous Blood

The vein blood (ca 0.3 mL) was collected in blood collection serum tubes coated with a clot activator on the inner wall of the tubes (Microvette® 200 Sarstedt, Marfour, Polska, cat. no. 20.1292). The blood test was carried out up to three hours after blood taking. For the biochemical profile, we used the Olympus AU680 Clinical Chemistry Analyzer and calorimetric method at 37 °C. We checked the following parameters and methods: enzymes (AST (aspartate aminotransferase), ALT (alanine aminotransferase)—IFCC Reference method with Pyrioxal Phosphate; Ap—IFCC Reference Method 2011), albumin—Bromocresol Green, urea—Urease/GLDH, creatinine—Jaffe IDMS and c-reactive protein.

Blood Collection and Plasma Preservation

Blood was collected immediately after euthanization with CO2 and transferred into Li-heparin tubes (Microvette 200; Sarstedt, Newton, NC, USA). Tubes were centrifuged at 2,000 × g for 10 min. Plasma was transferred into an Eppendorf tube, snap-frozen in liquid nitrogen, and kept at −80°C. Plasma (minimum 50 μL) biochemical analyses were done by the Division of Clinical Chemistry and Biochemistry at the University Children’s Hospital Zürich (Zürich, Switzerland) with a commercial routine method on an Alinity c (Abbot Laboratories, Chicago, IL, USA).
