Organoid Albumin and Bile Acid Assays

Functional studies were performed in collected supernatant or in whole organoids. To assess albumin production, culture medium was collected 1 week after the last medium change and albumin levels were assessed using an Albumin ELISA kit (Assay Pro) according to manufacturer’s instructions. Concentration of total bile acid was established using a Total Bile Assay kit (Cell Biolabs, inc.) according to manufacturer’s instructions on supernatant obtained after sonication of whole organoids in PBS.
