Lenti pac hiv qrt pcr titration kit

Manufactured by GeneCopoeia

The Lenti-Pac HIV qRT-PCR Titration Kit is a lab equipment product designed to quantify lentiviral vector titers. It utilizes quantitative reverse transcription-polymerase chain reaction (qRT-PCR) technology to accurately measure the concentration of lentiviral particles in a sample.

Lentiviral Transduction of Human Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Lentiviruses were produced in HEK293T cells using Lenti-Pac HIV Expression Packaging Kit, as per manufacturer’s guidelines (GeneCopoeia, Rockville, MD, USA; #LT001). Lentivirus titers were measured using Lenti-Pac HIV qRT-PCR Titration Kit (GeneCopoeia, #LT001) according to the manufacturer’s protocol. HFF cells were transduced with the different lentivirus preparations for 3 days in the culture medium supplemented with 5 µg/mL Polybrene (hexadimethrine bromide). Transduced cultures were either used immediately for downstream experiments or selected for 6 days using 2 µg/mL puromycin to generate stable cell lines. Plasmids, shRNA clones, and open reading frame expression clones were purchased from GeneCopoeia (Table S2).

Generating Lentiviral Vectors in 293FT Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The human embryonic kidney cells (293FT), cultured in 100-mm Petri dishes (Corning), were transfected with the lentiviral expression construct and other 3 packaging plasmids (psPAX2, pMD2.G, and pRSV-Rev) using X-treme GENE HP DNA Transfection Reagent (Roche Diagnostics GmbH, Germany), according to the manufacturer's protocols. The virus was collected 72 h after transfection and concentrated by ultracentrifugation. The viral titer was determined using the Lenti-Pac HIV qRT-PCR Titration Kit (GeneCopoeia), according to the manufacturer's protocols.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
