Genistein's Impact on HDAC Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
Nuclear extract was obtained from the untreated HeLa cells as mentioned in the previous section. Epiquik HDAC activity assay kit (Epigentek, New York, USA) was used to ascertain the effect of genistein (50 µM) on the activity of HDAC enzymes. Nuclear extract was added to the assay plate along with the buffers and genistein and placed at 37 °C for 1 h. Afterwards, ELISA based detection was performed to quantitate the product formed. The percentage of HDAC inhibition following genistein treatment in contrast to the untreated control was assessed by the following formula, where OD is optical density:

HDAC Activity Assay with Luteolin

Check if the same lab product or an alternative is used in the 5 most similar protocols
The EpiQuik HDAC activity assay kit (Catalogue number P-4002; Epigentek, NY, USA) was used to determine the effect of luteolin treatment on HDAC activity. Luteolin (5 µM, 10, and 20 µM), nuclear extract, and HDAC assay buffer were added to the assay plates and incubated for one hour at 37 °C to allow the action of the enzyme, followed by incubation with capture and detection antibody. In the controls, luteolin was substituted by assay buffer. The plate was read on an ELISA reader at 450 nm. Inhibition of HDAC activity by luteolin was calculated in comparison with control wells. Experiments were repeated thrice, and the mean ± SD was used to plot as a graph (p ≤ 0.05).

Assessing SFN's Impact on HDAC Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
The effect of SFN on HDAC activity in HeLa cells was determined using the EpiQuik HDAC activity assay kit (Epigentek, USA) according to the manufacturer's instructions. Briefly, HeLa cells were treated with SFN and TSA for 3 days and harvested and nuclear extracts were then prepared for the same using EpiQuik nuclear extraction kit (Epigentek, USA) following the manufacturer's instructions.

Chrysin's Modulation of HDAC Activity

Check if the same lab product or an alternative is used in the 5 most similar protocols
The EpiQuik HDAC Activity Assay Kit (Catalog Number P-4002, Epigentek, United States) was used for evaluating the effect of chrysin on the HDAC activity. The acetylated histone substrate–coated assay plate was prepared by adding 50 μL of the biotinylated HDAC substrate diluted in wash buffer to all wells. The assay plate was incubated at room temperature for 45 min and washed with wash buffer. HDAC assay buffer was dispensed to the wells in chrysin (5, 10, and 15 µM) treated and untreated nuclear extract placed. The plate was kept at 37°C for an hour followed by incubation with capture and detection antibody. Developing solution was used to develop the signal, stop solution stopped the reaction, and OD was read at 450 nm. The percentage inhibition of the HDAC activity against chrysin treatment was calculated by comparing with DMSO control. The experiment was repeated three times, and mean ± SD was used to plot a graph. One-way ANOVA was used to check the statistical significance at the p-value ≤ 0.05.

HDAC Activity Assay in Prefrontal Cortex

Check if the same lab product or an alternative is used in the 5 most similar protocols
The total HDAC activity was evaluated using the EpiQuik HDAC Activity Assay Kit (Epigentek, USA). Briefly, nuclear extracts of the PFC were obtained with the EpiQuik™ Nuclear Extraction Kit (Epigentek, USA), incubated with substrate and assay buffer at 37°C for 1 hr, followed by the incubation with a capture antibody at room temperature for 60 min. The detection antibody was incubated at room temperature for 30 min. The absorbance was read on a microplate spectrophotometer at 450 nm. The HDAC activity was measured according to the manufacturer's instructions.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
