K2edta coated microtainer tubes

Manufactured by BD
Sourced in United States

K2EDTA coated microtainer tubes are small, sterile containers used to collect and store small volumes of blood samples. The tubes are coated with the anticoagulant K2EDTA, which prevents the blood from clotting during collection and storage.

Circulating Neutrophil and Monocyte Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was obtained from mice and immediately added to K2EDTA-coated Microtainer tubes (BD, Franklin Lakes, NJ) to prevent clotting. The proportions of circulating neutrophils and monocytes within the white blood cell population was determined using the ADVIA® 120 Haematology System (Siemens Healthcare Diagnostics Inc, Tarrytown, NY).

At the time of euthanasia, mice were subjected to isoflurane overdose and blood was collected via cardiac puncture into K2EDTA coated microtainer tubes (BD Pharmingen, cat# 365967). Automated complete blood count with differential was performed using a Hemavet 950FS instrument (Drew Scientific).

Tumor and Immune Cell Isolation Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Prior to staining, tumors were weighed, mechanically dissociated, incubated in a shaker at 37°C for 30 minutes in RPMI-1640 medium containing 5% fetal bovine serum (FBS), 5 mg/mL collagenases IV and I (Gibco), and 40 U/mL DNase, and then passed through a 70-μm filter as a single-cell suspension. Spleens were crushed through a 70-μm filter and red cell lysis was performed with ammonium chloride–potassium (ACK) buffer (Gibco). Blood was collected by submandibular bleeding with a lancet into K2EDTA coated Microtainer tubes (BD Biosciences) followed by red cell lysis with ACK. Antibodies used for flow cytometry are listed in Supplemental Table 5. Cells were stained for cell surface expression in flat-bottom 96-well plates on ice in phosphate-buffered saline (PBS) with 2% FBS. Intracellular markers were stained using the eBioscience Foxp3/Transcription Factor Staining Buffer Set according to the manufacturer’s instructions. Fluorescently conjugated antibodies were used as per the manufacturers’ instructions. LIVE/DEAD Fixable Aqua Dead Cell Stain Kit (Thermo Fisher Scientific) was used to gate on live cells. Data were acquired on an Attune NxT Flow Cytometer (Thermo Fisher Scientific) and analyzed via FlowJo (Becton Dickinson). The gating strategy used for analysis is shown in Supplemental Table 6 and Supplemental Figure 5.

Blood samples were collected at experimental endpoints in K2EDTA-coated BD microtainer tubes (Becton, Dickinson and Company, Franklin Lakes, NJ, USA) for hematologic analyses followed by centrifugation at 1300 RCF for 10 min. Blood cell counts were performed on whole blood using a calibrated 950FS HemaVet apparatus (Drew Scientific, Waterbury, CT, USA) using the FS-Pak reagent kit, and white blood cell count (WBC) and differential leukocyte (%) count were measured.

Comprehensive Blood Cell Analysis

At the designated sacrifice time points, blood samples (500 μL) were collected in K2EDTA-coated BD microtainer tubes (Becton, Dickinson and Company, Franklin Lakes, NJ) and blood cell counts performed using a calibrated 950FS HemaVet apparatus (Drew Scientific, Waterbury CT). The blood samples were analyzed using the FS-Pak reagent kit and were measured for the following parameters: white blood cell count (WBC), differential leukocyte (%) count, hemoglobin concentration (HGB), hematocrit (HCT), red blood cell count (RBC), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MCH), mean corpuscular hemoglobin concentration (MCHC), red cell distribution width (RDW), platelet count (PLT), and mean platelet volume (MPV).

Postpartum Maternal-Pup Hematology

Check if the same lab product or an alternative is used in the 5 most similar protocols
At approximately one week postpartum, maternal and pup whole blood samples were collected into K2EDTA-coated BD Microtainer Tubes (BD, Franklin Lakes, NJ, USA; 365974) and stored on ice for a maximum of two hours until hematocrit and hemoglobin concentration values were assessed by an individual blinded to the cohort identities using a HESKA-HemaTrue® Veterinary Hematology Analyzer (Heska Corporation, Loveland, CO, USA; 5600). Within the analyzer, hematocrit values reflect the percentage of red blood cells in whole blood (%RBC). Hemoglobin concentrations reflect the number of grams of hemoglobin per deciliter of blood (g/dL), a value measured spectrophotometrically as RBC are lysed and intracellular hemoglobin is released within the analyzer. The trunk whole blood of pups was collected via decapitation, while maternal blood was collected utilizing a heparin-coated syringe (Sigma-Aldrich, St. Louis, MO, USA; H3149) for intravenous terminal blood draws through the inferior venae cava post ketamine overdose.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
