Establishing GFP-tagged H. pylori Infection in Hamsters

Two bacterial strains, cagA-positive and cagA-negative H. pylori tagged with green fluorescent protein mutant 2 gene (gfpmut2) were kindly provided by Prof. R. Haas, Max von Pettenkofer Institut für Hygiene und Medizinische Mikrobiologie [13 (link)]. Both strains were grown on GC selective agar plates, supplemented with 6 mg/L chloramphenicol (chloramphenicol supplement, Oxoid Limited, Hampshire, England), 10mL/L vitox (Vitox supplement, Oxoid Limited, Hampshire, England), Dent (H. pylori selective supplement, Oxoid Limited, Hampshire, England), and horse serum (5%). The plates were incubated for 4–5 days at 37 °C in 6–12% O2 and 5–8% CO2 microaerobic atmosphere adjusted by 1 sachet/2.5L jar of gas generator (AnaeroPack-MicroAero Gas Generator, Mitsubishi Gas Chemical Co. Ltd., Tokyo, Japan) [14 (link)]. After sufficient bacterial growth, H. pylori colonies were suspended in Brucella broth, mixed, and washed several times, and adjusted to 1.7 × 108 colony forming units (CFUs)/mL [15 (link)]. Hamsters were given 1 mL of the bacterial solution orally, using a stomach tube 24 h after fasting.

Bioaerosol and Dust Exposure in Plastic Waste Sorting Facilities

The study was conducted in 2 plastic waste sorting facilities in the Czech Republic. The analysis of the working environment was carried out in both facilities. Based on the analysis, a sampling place with the highest expected exposure of the employees to bioaerosol and dust was selected. The sampling place was next to the conveyor belt where the employees sort plastic waste. Samples were collected about 1.5 m above the ground. In both facilities, the samples were collected in 2013 and 2014 (October 2013, January 2014, May 2014). During agar base (Oxoid Ltd., UK) with the addition 100 mg/l of chloramphenicol supplement (Oxoid Ltd., UK); and YGC-medium ready-poured plate (Oxoid Ltd., UK). The cultivation media MEA, DRBC and SDA were prepared according to the manufacturer's instructions.
