Sourcing HCC Samples and Cell Lines

Check if the same lab product or an alternative is used in the 5 most similar protocols
HCC samples and paired adjacent tissues were obtained from surgery patients of Xijing Hospital, the Air Force Military Medical University, with informed consent in compliance with the relevant ethical regulations. Tissue microarrays containing 90 cases of HCC with paired adjacent tissues and follow-up information were purchased from Outdo Biotech Co., Ltd. (Shanghai, China). Human hepatoma cell lines SK-HEP-1, PLC/PRF/5, Huh-7, SNU-398, MHCC97L, Hep3B, HCCLM6, HepG2, and HEK293FT were purchased from the American Type Culture Collection (ATCC, VA) or obtained from the State Key Laboratory of Cancer Biology (CBSKL). Cells were cultured in DMEM supplemented with 10% fetal bovine serum and penicillin-streptomycin (100 U/mL) (Gcibco, USA) in an atmosphere with 5% CO2.

Culturing Human Liver Cell Lines

Check if the same lab product or an alternative is used in the 5 most similar protocols
The human liver cell line of L02 (normal), human HCC cell lines of Hep3B with low invasiveness, and highly invasive HLE, Huh7, BEL7402, SMCC7721, MHCC97L, MHCC97H, HCCLM3, and HCCLM6 cells were purchased from the American Type Culture Collection (ATCC, Manassas, VA, USA). All cell lines were grown routinely in Roswell Park Memorial Institute (RPMI) 1640 medium (Invitrogen, CA, USA) supplemented with 10% fetal bovine serum (FBS, Gibco, Carlsbad, CA, USA), 100 U/mL penicillin, and 100 μg/mL streptomycin (Invitrogen). Cells were cultured in a humidified incubator at 37°C with 5% CO2. The culture medium was replaced every 3 days. Cells were passaged as soon as the cell attachment rate reached 80%–90%.

Investigating Liver Cell Responses to Pharmacological Agents

Check if the same lab product or an alternative is used in the 5 most similar protocols
A normal human liver cell line (QSG‐7701) and liver cancer cells (Hep3B, MHCC97H, HepG2 and HCCLM6) were purchased from American Type Culture Collection (ATCC). The other cells were a gift from Dr. Qu and Dr. Chen. A normal human liver cell line (QSG‐7701) was cultured in RPMI‐1640 medium (HyClone), and cancer cell lines were cultured in Dulbecco's Modified Eagle Medium (DMEM) (HyClone) containing 10% foetal bovine serum (Biological Industries). Moreover, these cells were cultured in an incubator at 37°C with 5% CO2.
Necrostatin‐1, PD150606 and Z‐VAD‐FMK were obtained from Sigma Aldrich. The microtubule (MT)‐stabilizing agents epothilone B and paclitaxel and the MT‐destabilizing agents fosbretabulin and nocodazole were purchased from MedchemExpress Inc. Each of the reagents was dissolved in DMSO (Sigma Aldrich) and diluted to the appropriate concentrations.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
