All cells were cultured in RPMI-1640 medium (Welgene, Korea) containing 5% fetal bovine serum (Corning Costar, USA) and 1% antibiotic-antimycotic (Gibco, USA) in a 37°C incubator in an atmosphere of 5% CO2.

Celastrol Inhibits Cancer Cell Proliferation

The inhibition of cell proliferation by Celastrol was measured using an MTT assay. Celastrol was purchased from Cayman Chemical (Michigan, USA) and dissolved in dimethyl sulfoxide (DMSO). The human stomach adenocarcinoma cell line AGS and the gastric carcinoma cell lines MKN1, MKN45, SNU216, SNU-668, and YCC-2 were purchased from the Korea Cell Line Bank (KCLB, Seoul, Korea). The KCLB authenticates the phenotypes of these cell lines on a regular basis. The analysis of cell proliferation using MTT assay and migration using a woundhealing assay was performed as described previously (32) (link).

Cell cultures were maintained at 37°C in an atmosphere of 5% CO2. The phenotypes of these cell lines have been authenticated by the KCLB.
