The β-blockers were resuspended in dimethyl sulfoxide (DMSO). Stock solutions were stored at -20°C for Nebivolol (50 mM, Sigma-Aldrich, ref. #N1915) and Carvedilol (50 mM, Sigma-Aldrich, ref. #C3993) and at -80°C for Propranolol (150 mM, Selleckchem, ref. S4076). The antioxidants Mito-TEMPO (MT) and Troxerutin (TROX) were resuspended in DMSO and stock at -20°C. The solutions are diluted in culture medium ex-temporaneously for the experiments.

CAR-T Cells Coculture with M2 Macrophages

Check if the same lab product or an alternative is used in the 5 most similar protocols
The 5×105 THP-1 cells/well or 1×106 PBMCs were seeded into the six-well plates and polarized into M2 macrophages. After thoroughly washing to remove all PMA and cytokines, the macrophages were cocultured with CAR-T cells and Raji cells. The ratio of CAR-T cells, tumor cells, and macrophages was 1:3:1. In some experiments, the cells were treated with thalidomide (1 µM), VX-765 (20 µM), Ac-YVAD-CMK (10 µM), prazosin (10 µM), propranolol (10 µM), or epinephrine (1 µM) (Selleck Chemicals).

Preparation of Chemotherapeutic Drug Solutions

Check if the same lab product or an alternative is used in the 5 most similar protocols
Doxorubicin and propranolol were purchased from Selleckchem. Stock solutions were prepared at 10 mM and 50 mM in dimethylsulfoxide (DMSO) and stored in aliquots at − 20 °C and − 80 °C, respectively. The chemotherapeutic drugs Vinorelbine (Vinorelbine Injection 50 mg/5 ml-Sandoz) and Docetaxel (Docetaxel Accord 160 mg/8 ml-Accord) were supplied by the Institutional Pharmacy Unit. Further dilutions were made in culture medium.

Selective Beta-Adrenergic Receptor Modulators

Check if the same lab product or an alternative is used in the 5 most similar protocols
Bisoprolol and propranolol were purchased from Selleckchem (Planegg, Germany). ICI‐118,551 was obtained from Sigma‐Aldrich (St Louis, MO, USA). Dimethyl sulfoxide (DMSO), obtained from MP Biomedicals (Illkirch Graffenstaden, France), was used as the solvent for Bisoprolol and propranolol, whereas ICI‐118,551 was dissolved in sterile H2O. Bortezomib was purchased from Selleckchem and melphalan from Sigma‐Aldrich; both were dissolved in DMSO following the manufacturer's instructions. 3‐Methyladenine (3MA), obtained from Selleckchem, was dissolved in sterile H2O following the manufacturer's instructions.

Isolation and Characterization of MDSCs

Check if the same lab product or an alternative is used in the 5 most similar protocols
PMN-MDSCs and M-MDSCs in the mouse spleens under the indicated conditions were FACS-sorted on the BD FACSAria. The cells were in vitro cultured in RPMI-1640 (Gibco) supplemented with 10% HI-FBS, 100 U/mL penicillin and 100 μg/mL streptomycin, and then treated with 10 μM NE or 20 μM propranolol (Selleck Chemicals) for 1 h. Total RNAs of the cells were extracted by the RNeasy Mini Kit and analyzed by the SYBR Green Real-Time PCR Kit (Thermo Fisher Scientific). Gapdh mRNA levels were utilized as the internal control.

NSCLC Cell Line Culture and Drug Preparation

Check if the same lab product or an alternative is used in the 5 most similar protocols
Human NSCLC cell lines H522, H460, A549, and H292 were purchased from the American Type Culture Collection (ATCC). All cell lines were cultured in RPMI 1640 medium (Gibco) containing 10% fetal bovine serum (Gibco) at 37 °C in a 5% CO2 incubator. Apatinib (Selleckchem) and anlotinib (Selleckchem) were dissolved in dimethylsulfoxide (DMSO) and stored at −80 °C (20 mM, stock solution). Propranolol (Selleckchem) was dissolved in DMSO and stored at −80 °C (50 mM, stock solution). KG501 (Selleckchem) was dissolved in DMSO and stored at −80 °C (25 mM, stock solution).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
