Generating ALKO Mice: GFAP-cre and LepR-floxed

To generate ALKO mice, we used a cre recombinase transgenic mouse under the control of a mouse glial fibrillary acidic protein (GFAP) promoter. The GFAP-cre mice were B6.Cg-Tg(Gfap-cre)73.12Mvs/J (Stock number 012886, Jackson Laboratory, Bar Harbor, MN) that contain a mouse GFAP promoter sequence directing expression of Cre recombinase (Garcia et al., 2004 (link)). The LepR-floxed mice were B6.129P2-Leprtm1Rck/J (Stock number 008327, Jackson Laboratory), with loxP sites on either side of exon 1 (Cohen et al., 2001 (link)). GFAPcre/+ hemizygous transgenic mice and LepRloxP/loxP homozygote mice were bred for two generations. The hemizygotes of the F1 generation were backcrossed with LepR-floxed mice, to select the F2 generation of ALKO-Δ1 mice that express both GFAP-cre and LepR-floxed alleles on tail genotyping. The agarose gel electrophoresis of PCR products and genotyping primers are shown in figure 1A and table 1.

Transgenic Mice for Obesity Research

This study was approved by the institutional committee of Osaka University. C57BL/6 J mice were obtained from Charles River Japan or Japan SLC. B6N.Cg-Tg (Pdgfra-cre/ERT) 467Dbe/J (stock no. 018280) and B6.129P2-Leprtm1Rck/J (stock no. 008327) were purchased from the Jackson Laboratory. The experiments were performed in accordance with the Guide for the Care and Use of Laboratory Animals of the Graduate School of Medicine Osaka University (no. 24-067-055).

Genetic Lineage Tracing of Mice

All mice were maintained on a C57BL/6 genetic background. The Leprflox/flox line (B6.129P2-Leprtm1Rck/J, stock no. 008327 from the Jackson Laboratory,) was kindly provided by L. Ma (Fudan University). The Shh-Cre line [42 (link)] (Stock No. 005622 from the Jackson Laboratory) and Nestin-Cre line (B6.Cg-Tg (Nes-cre)1Kln/JNju, Stock No. J003771 from Model Animal Research Center of Nanjing University) were kindly provided by Z. Yang (Fudan University). All mice were housed under a 12-h light/dark cycle and had ad libitum access to food and water in a controlled animal facility. All animals were treated in accordance with protocols approved by the Animal Care and Use Committee of Shanghai Medical College of Fudan University.
