Immunohistochemistry for MMR Proteins

IHC for four MMR proteins (MLH1, PMS2, MSH2 and MSH6) was performed on formalin‐fixed, paraffin‐embedded tissue obtained from representative sections of the resection specimens. MLH-1 (Ventana 790–5091), PMS2 (Ventana 790–5094), MSH2 (Ventana 790–5093), and MSH6 (Ventana 790–5092) were performed according to previously validated protocols using the Ventana auto-staining platform (Roche). Normal colonic crypt epithelium adjacent to the tumor, lymphoid cells and stromal cells served as internal positive controls. The cases with loss of any of MMR protein expression were defined as dMMR, and the cases without loss of MMR protein expression were defined as pMMR (Supplement Fig).
