Profiling Pro-Inflammatory Factors in Mice

The levels of the following pro-inflammatory factors were measured: tumor necrosis factor-α (TNF-α; cat no. PT512; Beyotime Biotechnology), interleukin (IL)-2 (cat no. PI575; Beyotime Biotechnology), IL-4 (cat no. PI612; Beyotime Biotechnology), IL-5 (cat no. PI620; Beyotime Biotechnology), IL-13 (cat no. PI627; Beyotime Biotechnology), IL-17 (cat no. PI545; Beyotime Biotechnology), interferon-γ (IFN-γ; cat no. PI508; Beyotime Biotechnology), OVA-specific IgE (cat. no. 439807-1; BioLegend, Inc.) and leukotriene C4 (LTC4; cat. no. E-EL-M0753 km−1; Shanghai Zhenyu Chemical Technology Co., Ltd). The measurement was performed in mice of different groups by ELISA specific kits according to the manufacturer’s instructions. Each experiment was repeated at least three times.

Anti-inflammatory Effects of CS-Gel Hydrogel

Lipopolysaccharide (LPS), silver nitrate (AgNO3), CS (degree of deacetylation ≥95%; Aladdin, CAS:9012-76-4), Gel (from bovine skin; Solarbio, CAS:9000-70-8) and TA were purchased from Sigma-Aldrich, USA. Antibodies specific for p65 (4764), α-tubulin (2144) and lamin B1 (17416) were purchased from Cell Signaling Technology (Danvers, MA, USA). Reactive Oxygen Species Assay Kit (S0033), interferon-γ (IFN-γ) (P6137), ATP (ST1092), nuclear and cytoplasmic protein extraction kit (P0027), IL-1β enzyme-linked immunosorbent assay (ELISA) kit (PI305), TNFα ELISA kit (PT512) and IL-6 ELISA kit (PI326) were purchased from Beyotime Biotechnology (Shanghai, China). All other solvents used were of analytical reagent grade.

Realgar-Based Anticancer Protocol

Highly purified realgar supplied by the Sanmenxia Yuhuangshan Pharmaceutical Co., Ltd was prepared from mined natural realgar. The purity of As4S4 in the realgar preparation was greater than 95.0%, confirmed by repeated X‐ray powder diffraction analyses. Realgar was dissolved in Dulbecco's phosphate‐buffered saline (DPBS) (Thermo Fisher Scientific) and sterilized by filtration. The content of As in DPBS solution was determined by inductively coupled plasma atomic emission spectrometry at the Instrumental Analysis Center of Shanghai Jiao Tong University (Shanghai, People's Republic of China). As4S4 stock solution of 766.36 μM was stored at 4°C. DDP was supplied by Qilu Pharmaceutical. Anti‐p53 (cat: 21891‐1‐AP), anti‐PD‐L1 (cat: 17952‐1‐AP), and anti‐GAPDH (cat: 10494‐1‐AP) antibodies were purchased from Proteintech. Pifithrin‐α (PFT‐α) (cat: S1816‐25 mg), methyl‐thiazolyl‐tetrazolium (MTT) (cat: ST316), interferon‐γ (IFN‐γ) (cat: P5664‐100 ug) and sodium dodecyl sulfate (SDS) powder (cat: ST626) were purchased from Beyotime Institute of Biotechnology. MTT powder was dissolved in DPBS at a concentration of 5 mg/ml. SDS powder was dissolved in water at aconcentration of 10%.
