Microbiota Depletion in Mice

For specific deletion of colon microbiota, mice were treated by enema with 500 µl ABX solution containing neomycin (#MB1716, Meilunbio, 2 g L−1), vancomycin (#MB1260, Meilunbio, 1 g L−1), and imipenem (#MB1748, Meilunbio, 0.5 g L−1) twice a day for 2 weeks.
For depletion of microbiota along the whole gut, mice were treated with an ABX solution containing neomycin (#MB1716, Meilunbio, 1 g L−1), vancomycin (#MB1260, Meilunbio, 0.5 g L−1), and imipenem (#MB1748, Meilunbio, 0.25 g L−1). ABX was dissolved into the sterile drinking water. Solutions and bottles were changed twice a week. Mice were treated for two weeks before tumor implantation and were continuously treated throughout the experiment.

Imipenem-Cilastatin Pharmacokinetic Study

Screening Antibiotic Resistance Genes and Potential Inhibitors

The bacterial strains, plasmids and primers used in this study are listed in Supplementary Table S1, Table S2 and Table S3, respectively. All bacterial strains were grown on Luria-Bertani (LB) plates or in LB broth. All NDM-1-positive strains were originated from our previous studies [17] . E. cloacae 20710 and E. cloacae 20712 were provided by Dr. Yonghong Xiao at Zhejiang University [18] . A. baumannii 21 was provided by Dr. Zhimin Guo at the First Hospital of Jilin University.
The full gene sequences of blaIMP-1, blaVIM-1, blaKPC-2 and blaOXA-48 were synthesized by GeneScript (Nanjing, China) and were inserted into pET28a or pETSUMO for protein expression.
The FDA-approved drug library was purchased from ApexBio Technology (Cat#L1021, Houston, TX, USA). Dexrazoxane, embelin, CAN and NDGA were dissolved in DMSO (Sigma-Aldrich, St. Louis, MO, USA). Meropenem, amoxicillin, ciprofloxacin, imipenem, erythromycin, chloramphenicol, tetracycline and gentamicin were obtained from Dalian Meilun Biotechnology Co., Ltd. (Dalian, China).

Antimicrobial Susceptibility Testing Protocol

Susceptibility to antimicrobials was determined in triplicate using the standard agar dilution method recommended by the Clinical and Laboratory Standards Institute (CLSI, 2020) [66 ]. The following eighteen antimicrobials were tested: kanamycin (KAN), gentamicin (GEN), erythromycin (ERY), azithromycin (AZM), norfloxacin (NOR), ofloxacin (OFX), ciprofloxacin (CIP), lomefloxacin (LOM), levofloxacin (LEV), sulfadiazine (SD), trimethoprim (TMP), ceftriaxone (CRO), cefixime (CFM), ampicillin (AMP), amoxicillin (AML), aztreonam (ATM), imipenem (IPM) and tetracycline (TET) (Meilun Biotechnology Co., LTD, Dalian, China). The isolates were grown on TSA plates, suspended in stroke-physiological saline solution to a turbidity equivalent to 0.5 McFarland Standard, and inoculated onto Mueller-Hinton agar plates using a multipoint inoculator (MIT-60P; Sakuma Seisakusyo, Tokyo, Japan). The final inoculum was approximately 104 CFU per spot. Plates were incubated at 37 °C for 16–18 h. The range of 2-fold concentrations used to determine the susceptibility was determined by CLSI criteria. In addition, the results were interpreted in accordance with CLSI criteria (Supplementary Table S3). E. coli ATCC25922 and E. faecalis ATCC29212 were used as quality control strains.
