Corall kit

Manufactured by Lexogen

The Corall kit is a RNA library preparation solution for highly accurate and efficient 3'-end sequencing. It enables the construction of 3'-end-enriched libraries from small amounts of total RNA or poly(A)+ RNA samples.

Poly-A mRNA Purification and Sequencing

The Dynabeads mRNA purification kit (Thermo Fisher Scientific) was used to extract all RNA molecules with a poly-A tail. An amount of 1–2 µg total RNA was used as input. Eluted RNA was used as input using the Corall kit (Lexogen, Vienna, Austria) according to the manufacturer’s instructions and targeting an insert size of 350 bp.

RNA-seq of microbial communities

RNA samples were delivered to Core Unit SysMed (Julius-Maximilian-University Würzburg) for RNA-seq. Briefly, after treating RNA samples with DNase, sufficient RNA quality was verified on a bioanalyzer (RNA chip, Agilent). Ribosomal RNA was depleted (RiboCop-META kit, Lexogen), and RNA was then reverse transcribed for cDNA library preparation using the CORALL kit (Lexogen) according to the manufacturer's instructions. After pooling cDNA library samples at equimolar amounts, quality was verified using a bioanalyzer (DNA chip, Agilent). The cDNA pools were sequenced using the NextSeq 500 system (HighOutput flow cell, 400 M, 1× 75 cycle single-end; Illumina).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
