Quantification of Veterinary Antibiotics in Samples

Standards of five veterinary antibiotics (VAs) (Figure 3) (Tetracycline (TC, 90%), OxyTetracycline (OTC, 98%), Sulfadiazine (SDZ, 98%), Norfloxacin (Norf, 98%), Enrofloxacin (Enorf, 98%)) were purchased from Sigma-Aldrich (St. Louis, MO, US) (CAS Reg. No. 60-54-8 for TC, CAS Reg. No. 6153-64-6 for OTC, CAS Reg. No. 68-35-9 for SDZ, CAS Reg. No. 70458-96-7 for Norf, and CAS Reg. No. 93106-60-6 for Enorf) (the structures are presented in Figure 4). Acetonitrile (ACN) and methanol (MeOH) of HPLC grade were purchased from Sigma-Aldrich. Oxalic acid, disodium ethylene–diaminetetraacetat (Na2EDTA), disodium hydrogen phosphate (Na2HPO4), and sodium dihydrogen phosphate (NaH2PO4) of analytical-reagent grade were obtained from Beijing Chemicals Company (Beijing, China). Deionized water (Milli-QMillipore, Bedford, MA, USA) was used in the study.

Antibiotic Resistance Profiling of Yeast

Antibiotic-resistance was carried out with the Kirbye Bauer method according to the NCCLS protocol [53 ]. Five different antibiotics were used: streptomycin (ST), chloramphenicol (CH), gentamicin (GE), erythromycin (ER), and oxytetracycline (OX) (Sigma, Milan, Italy). Tests were performed on YPD agar added with five different concentrations of each antibiotic: 40, 80, 100, 130 and 150 µg/mL, in comparison to the control without antibiotic addition. Cell suspensions were adjusted to 0.2 of Optical Density (OD) (106 cells/mL), and 8 µL of each strain were dropped on the antibiotic-YPD plates, which were incubated at 37 ± 2 °C for 48 h. Cell survival and growth level on the different concentrations of the five antibiotics were qualitatively evaluated, by comparing the size and thickness of each strain spot with the corresponding control. All the experiments were performed in duplicate.

Standards and Reagents for Tetracycline Assays

The standards of tetracycline (TC), chlortetracycline (CTC), minocycline (MC), doxycycline (DC), oxytetracycline (OTC), demeclocycline (DMC), and tigecycline (TIC) were from Sigma-Aldrich (St. Louis, USA). Methacycline (MTC) was from J&K Scientific Ltd. (Beijing, China). Lymecycline (LMC) was from Toronto Research Chemicals (Toronto, Canada). Sancycline (SC), fluoresceinamine (FA), and fluorescein isothiocyanate (FITC) were purchased from Shanghai Yuanye Biological Technology Co., Ltd. (Shanghai, China). All the biological reagents (isopropyl-beta-D-thiogalactopyranoside, culture medium, express vector pET32a) and the kits (EasyPure Quick Gel Extraction Kit, Fast MultiSite Mutagenesis System, sodium dodecyl sulfate polyacrylamide gel electrophoresis (SDS-PAGE) gel) were the same as our recent report [29 (link)].

Tetracycline-Based Magnetic Biochar Synthesis

Doubly deionized water (DDW), obtained using WaterPro Water System (Labconco Corp., Kansas City, MO, USA), was used throughout the experiments.

Seedling Treatment Reagent Acquisition

Antibiotic Susceptibility Evaluation Protocol

For this study, Sigma-Aldrich (St. Louis, MO, USA) provided oxytetracycline of high purity (98%). Inorganic salt medium (ISM) was comprised of—(NH4)2SO4 (2.0 g), K2HPO4 (0.5 g), and NaH2PO4 (0.5 g) adjusted to pH 7.0. Growth media—nutrient broth and nutrient agar, Mueller–Hinton agar, and OTC (g/l) constituted of (NH4)2SO4 (2.0 g), K2HPO4 (0.5 g), and NaH2PO4 (0.5 g). The solid medium contained 20 grams of agar per litre. Hi-Media provided commercial-grade antibiotic susceptibility discs for six antibiotics: tetracycline, gentamicin, ceftriaxone, oxytetracycline, erythromycin, and vancomycin. Merck provided acetonitrile and methanol of high-performance liquid chromatography grade.

Comprehensive Veterinary Drug Extraction

The Barnstead NANOpure™ DIamond (Waltham, MA) purification system was used to obtain 18.2 MΩ water. LC-MS grade methanol and acetonitrile were obtained from EMD Millipore Corporation (Billerica, MA), and formic acid (88%) was purchased from Fisher Chemical (Pittsburgh, PA). Oasis™ HLB solid-phase extraction (SPE) cartridges were purchased from Waters (Milford, MA).

Quantification of Antibiotics and Pesticides

Antibiotics and pesticide residues were analyzed using analytic grade reagents. Standards of high purity grades (>99%) for 26 antibiotics belonging to four families: Sulfonamides (sulfacetamide, sulfisomidine, SMT, sulfapyridine, sulfamerazine, sulfameter, sulfachloropyridazine, sulfamethoxazole, sulfabenzamide, sulfadimethoxine, sulfaquinoxaline, sulfamethoxypyridazine, and sulfadoxine), quinolones (nalidixic acid, oxolinic acid, CIP, norfloxacin, ofloxacin, flumequine, danofloxacin, enrofloxacin, lomefloxacin, and sarafloxacin), tetracyclines (tetracycline and oxytetracycline), and β-lactams (ampicillin) in addition to a hundred pesticide standards were procured from Sigma-Aldrich. Each pesticide stock solution was accurately prepared in acetonitrile at a concentration of 1 g/L. The working standards were freshly prepared by diluting the stock solution in acetonitrile to the desired concentration. Both stock and working solutions were stored at −20°C.
High-performance liquid chromatography (HPLC)-grade water, HPLC-grade acetonitrile, disodium ethylenediaminetetraacetate dihydrate (Na2 EDTA), and magnesium sulfate (MgSO4) were also supplied by Sigma-Aldrich.
