HPLC Analysis of PhIP in Samples

All solvents used in this study were of HPLC grade. Acetonitrile, methanol, phosphoric acid, and trimethylamine (99.5%) were purchased from Merck (Darmstadt, Germany). Glycerol, D(+)-glucose, L-proline (99%), glycine (99%), l-phenylalanine (99%), and creatinine were obtained from R&M (R&M Marketing, Essex, U.K). 2-amino-1-methyl-6-phenylimidazo[4,5-b]pyridine (PhIP) was obtained from Toronto Research Chemicals (Toronto, Canada). A stock solution of 100 ppm PhIP standard was prepared in methanol and kept at 4 °C.

Cinchona Alkaloid Quantification by SFC

Six Cinchona alkaloids (compounds 1-6, see Fig. 1 for structures) with a purity ≥98% were available as standards; they were purchased from Phytolab (Vestenbergsreuth, Germany; compounds 1 and 2) and Sigma Aldrich (St. Louis, MO, USA; compounds 3-6). Plant samples (CC-2017-1 to CC-2017-4) were bought 2017 in different pharmacies in Innsbruck, Austria; voucher specimens are deposited at the Institute of Pharmacy, Pharmacognosy, University of Innsbruck. Compressed carbon dioxide for SFC analysis had a purity of ≥99.995% (4.5 grade) and came from Messer (Gumpoldskirchen, Austria). All solvents and reagents (methanol, acetonitrile, diethylamine, trimethylamine, sodium hydroxide, acetic acid, ammonium acetate, phosphoric acid) utilized in this study were of analytical grade and purchased from Merck (Darmstadt, Germany). An Arium 611 water purification system from Sartorius (Göttingen, Germany) produced the required HPLC grade water.

Metabolite Extraction and Analysis

HPLC Quantification of Microbial GABA

GABA produced after fermentation was measured using HPLC (Agilent Tech, Waldron, Germany). The HPLC had Hypersil ODS C18 reverse-phase column with 250 mm length, 5 μm diameter, and 4.6 mm width. The culture broth (100 μL) was sieved using a 0.22 μm filter. Mobile phase A of the HPLC was filled with 10.254 g of 99% sodium acetate three hydrates (Sigma-Aldrich, USA). It was then dissolved in 900 mL of deionized water and 500 μL trimethylamine (Merck). Glacial acetic was used to adjust the pH of the mobile phase A to 5.8. Acetonitrile (HPLC grade, Merck) and deionized water were the mobile phases B and C, respectively. Phases were filtered through 0.22 μm membrane filter, with an injection volume of 20 μL. The final compound was identified by the detector at 254 nm. The quantity of GABA produced was computed by comparing the peak to the GABA standard.

Poloxamer-188 Formulation for Diltiazem Delivery

Penicillin-streptomycin antibiotic cocktail was purchased from Hi-Media (India), isoflurane (Forane) from Abbott (India), Ceftriaxone (Cefaxone) from Lupin (India), Tramadol (Dolphan) from GreenCross Remedies (India) and povidine-iodine (Betadine) from Win-Medicare (India). Nano-hydroxyapatite (HA) used was synthesized in the laboratory using the protocol as described elsewhere 51 (link) .

Radiolabeling of [18F]Fallypride Precursor
