Polymer-based Periodontal Membrane Fabrication

Antibacterial Sutures for Wound Healing

Violet colored polydioxanone (PDS II®, Ethicon Inc., Somerville, NJ) monofilament surgical sutures were used. 1,1,1,3,3,3-hexafluoro-2-propanol (HFP, Sigma-Aldrich, St. Louis, MO) was purchased and employed as a solvent for PDS II®. Therapeutic agents (i.e., metronidazole and ciprofloxacin) were obtained from Sigma-Aldrich. The distinct bacteria, namely Lc (ATCC 4646), Ss (ATCC 25975), Fn (ATCC 10953) and Aa (ATCC 33384) were obtained from American Type Culture Collection (ATCC). Tryptic Soy Broth (TSB) culture media, Rogosa agar and Columbia Blood Agar base were obtained from Difco (Sparks, MD, USA). Sucrose, phosphate buffer saline (PBS), sodium chloride and paraformaldehyde were obtained from Fisher Scientific (Fair Lawn, NJ, USA). Defibrinated sheep blood was obtained from Carolina Biological Supply (Burlington, NC, USA). Hexamethyldisilazane (HMDS) was obtained from Sigma-Aldrich. All materials and chemicals were used as-received without any additional purification.

Hybrid Collagen-PCL Scaffold Fabrication

Peri-implantitis Bacterial Cultures and Polymer Synthesis

Type-B Gelatin (GEL) from bovine skin (bloom no. 225), tetracycline hydrochloride (TCH, molecular weight [Mw] 480.90 g/mol), hexamethyldisilazane (HMDS), formaldehyde, and 1,1,1,3,3,3-hexafluoro-2-propanol (HFP) were acquired from Sigma-Aldrich (Sigma-Aldrich Corporation, St. Louis, MO, USA). Brain heart infusion (BHI, Difco Laboratories Inc., Detroit, MI, USA) culture media containing 5 g/L yeast extract (BHI-YE, Difco Laboratories Inc., Detroit, MI, USA) with 5% v/v Vitamin K/hemin (Thermo Fisher Scientific, Inc., Pittsburgh, PA, USA) solution was used. Phosphate-buffered saline, ethanol, and blood agar plates supplemented with 5% sheep blood were obtained from Fisher Scientific Company LLC (Fair Lawn, NJ, USA).

Collagen-based Polymer Scaffold Synthesis

Osteogenic Potential of Ca-TMP Biomaterial

We obtained 1,1,1,3,3,3-hexafluoro-2-propanol (HFP) from Sigma-Aldrich in St. Louis, MO, USA. The calcium trimetaphosphate (Ca-TMP) used had a molecular weight of 594.07 and a particle size of 236 ± 50 nm. It was synthesized in the Laboratory of Pediatric Dentistry of the Department of Preventive and Restorative Dentistry at São Paulo State University (FOA/UNESP). The alpha-minimum essential medium (α-MEM), fetal bovine serum (FBS), and penicillin/streptomycin were procured from Gibco in Grand Island, NY, USA. Invitrogen in Waltham, MA, USA, provided AlamarBlue® (DAL1100), ActinRed™ 555, and ActinGreen™ 488. Additionally, we used the SensoLyte p-nitrophenyl phosphate (pNPP) ALP kit from AnaSpec, Inc. in Fremont, CA, USA.

Doxorubicin, ECL substrate kit, Protease inhibitor cocktail, and Phosphatase inhibitor cocktail were obtained from Abcam (Cambridge, UK). Dulbecco’s Modified Eagle Medium (DMEM) and Trypsin EDTA 0.05% were procured from Thermo Fisher Scientific (Waltham, MA, USA). 10X RIPA, Fetal bovine serum (FBS), and Phosphate Buffer Saline (PBS) were purchased from Himedia (Mumbai, India). Other standard laboratory chemicals and reagents were purchased from Sisco Research Laboratories (SRL) Pvt. Ltd, India.
