Electrophysiological Recordings of Dark-Adapted Murine Retinas

Full field ERGs were recorded in dark-adapted (18 h) mice (Rpe65-450Met, agouti) at the indicated ages. The mice were anaesthetized with an intraperitoneal injection of a mixture of ketamine (80 mg/kg), xylazine (10 mg/kg), and acepromazine (5 mg/kg). Pupils were dilated with topical phenylephrine hydrochloride and tropicamide and body temperature was maintained at 35 to 37 °C. Recordings were acquired with a Celeris rodent ERG system (Diagnosys) and a pair of corneal electrodes positioned with GenTeal Tears gel (Alcon Laboratories). Scotopic ERG responses in dark-adapted mice were elicited by single flashes having intensities from 0.01 cd·s/m2 to 100 cd·s/m2. Photopic responses from 0.1 to 100 cd⋅s/m2 intensities were recorded in mice adapted to background light of 30 cd⋅s/m2 (10 min). Triplicate responses were computer-averaged for each flash condition. The amplitude of the a-wave was measured from the baseline to the first trough. The amplitude of the b-wave was measured from the trough of the a-wave to the following positive peak.

Photopic ERG Measurement in Rho-/- Mice

ERG was carried out as previously described (Yu et al., 2020). The mice were dark-adapted for 12 hours. The experiments were performed in the dark with limited 589 nm red light illumination. Mice were anesthetized with an intraperitoneal injection of ketamine (100 mg/kg) and xylazine (20 mg/kg). Tropicamide 0.5% (Sandoz, West Princeton, NJ, USA) was used for pupil dilatation. Mice were placed on a heating pad to maintain body temperature during testing and prevent stress cataract formation. Contact electrodes were placed centrally on both corneas. The electrodes were lubricated with eye gel (GenTeal Tears gel; Alcon, Geneva, Switzerland), the ground electrode was placed subcutaneously at the base of the tail, and a reference electrode was inserted subcutaneously at the forehead between the eyes. Rho–/– mice generally lack scotopic responses after 7 weeks of age; a photopic ERG was recorded using ColorDome LabCradle (Diagnosys, Lowell, MA, USA) in Rho–/– mice and after tcES/sham treatments.

Optical Coherence Tomography Imaging of Mouse Retina

WT and F3−/− animals were anesthetized using ketamine/xylazine (100/10 mg/kg). Once unresponsive to foot pitch, the mouse was placed in a stage for SD-OCT (Bioptigen Envisu R2210; Morrisville, NC, USA). One drop of 1% tropicamide (Sandoz #61314–0355-02, West Princeton, NJ, USA) was placed on each eye to dilate the pupils, and GenTeal Tears gel (Alcon Laboratories Inc. # AL042947; Fort Worth, TX, USA) was applied to prevent the corneal surface from drying. The optic disc was located manually and was used to center the scans. The retina was scanned at 1.8 mm × 1.8 mm 1000 × 100 × 10 and averaged to 1000 × 100 × 1. Images were analyzed using the Diver Software.
