Mouse albumin elisa quantitation kit

Manufactured by Fortis Life Sciences
Sourced in United States

The Mouse Albumin ELISA Quantitation Kit is a laboratory tool used to measure the concentration of mouse albumin in various biological samples. It utilizes the enzyme-linked immunosorbent assay (ELISA) principle to quantify the target protein. The kit includes all the necessary components to perform the assay, including pre-coated plates, standards, and reagents.

Equal volumes of urine samples were examined by SDS-PAGE followed by Coomassie Brilliant Blue (Sigma-Aldrich) staining. Urine albumin concentrations were assayed using a mouse albumin ELISA quantitation kit (Bethyl Laboratories Inc). Urine and serum creatinine levels were measured by a creatinine assay kit (BioAssay Systems) according to the manufacturer’s instructions. Albumin-to-creatinine ratios were calculated to assess the severity of albuminuria.

To assess albumin secretion, media samples were collected at different time points. After omitting cross-reactivity for bovine albumin, the amount of secreted albumin was determined using a mouse albumin ELISA quantitation kit or a human albumin ELISA quantitation kit (Bethyl laboratories, Inc., UK) and normalized for the amount of cells using an MTT assay.

Measuring Lung Inflammation Markers

Protein concentration and cellularity of BAL fluid was measured as described previously18 (link),26 (link). Briefly, 0.7 ml of sterile PBS was instilled into mouse lungs, lavaged twice, collected and centrifuged. Total protein concentration of the supernatant was measured using a Bradford assay (Bio-Rad), and the total number of cells was determined using a hemocytometer and then cells were stained with Diff-Quick (Siemens, Newark, DE, USA) after cytospin. Albumin levels in BAL fluid of each group was assessed using a mouse albumin ELISA quantitation kit (Bethyl Laboratories, Montgomery, TX) as described18 (link).

Creatinine in urine and blood was determined using a colorimetric assay (ABX Pentra Creatinine 120 CP kit, Horiba ABX SAS, Montpellier, France) according to the manufacturer’s instructions. Na+ and K+ were determined by flame photometry (model IL 943, Instrumentation Laboratory, Lexington, MA). Albumin was measured according to the manufacturer’s instructions using the Mouse Albumin ELISA Quantitation kit (Bethyl Laboratories, Inc., Montgomery, USA).

Hepatocyte conditioned media (CM) were collected on
days 3 and 5 from the different groups. We evaluated Alb
production using a Mouse Albumin ELISA Quantitation Kit
(Bethyl Laboratories, USA) and urea secretion with a Urea
Assay Kit (Biorex, UK). The values were normalized to the
total protein acquired from a Total Protein Kit (Biorex, UK)
that used an auto analyzer (Alpha-Classic, Iran).

Quantifying Urinary Albumin and Creatinine

Check if the same lab product or an alternative is used in the 5 most similar protocols
Urine albumin levels were measured using mouse albumin ELISA quantitation kit (Bethyl Laboratories, Montgomery, TX, USA). Urine creatinine was determined by a creatinine assay kit (BioAssay Systems, Hayward, CA, USA). To rule out the influence of glomerular filtration rate on urinary excretion of albumin, urine albumin levels were adjusted by urine creatinine levels and presented as urine albumin-to-creatinine ratios.

Murine Urinary Albumin and Creatinine Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
Urine from the mice was collected into siliconized tubes. For the albumin assay, the urine was diluted 1:1,000 and measured in duplicate with the Mouse Albumin ELISA Quantitation Kit (Bethyl Laboratories Inc., USA). Optical density values were obtained using the MRX microplate reader (Dynex Technologies Inc., USA) at a wavelength of 450 nm. Albumin values (ng/ml) were determined as the mean of duplicate values. The urine creatinine (mg/dl) was measured using the alkaline picrate (Jaffé) method. Creatinine LiquiColor Test was purchase from StanBio Lab Inc. (USA). Urine creatinine assay was performed on spectrophotometer (Bio-Rad, capable of absorbance readings at 510 nm) in accordance with the manual steps. The urinary albumin/creatinine ratio (mg/g) was calculated for each animal.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
