Antibody Detection of Signaling Proteins

The antibodies (GRK4 and D1R) were rabbit anti‐rat antibodies and were purchased from Santa Cruz Biotechnology, Inc. (Dallas, TX) and Millipore (Billerica, MA). The phosphoserine antibody was a rabbit anti‐rat antibody and was purchased from Cell Signaling Technology (Beverly, MA). Fenoldopam, a D1‐like receptor agonist, was purchased from Sigma (St. Louis, MO). All other chemicals for the various buffers were of the highest purity available and purchased from Sigma or Gibco (Grand Island, NY).

In Vivo and In Vitro Endotoxemia Models

LPS (E. coli 0111:B4), capsaicin, reserpine, fusaric acid, dopamine, fenoldopam, pergolide and butaclamol were all purchased from Sigma-Aldrich® (Saint Louis, MO) and dissolved in sterile, pyrogen-free PBS (Gibco®, Life Technologies, Grand Island, NY). capsaicin (5 mg kg−1 i.m.; dissolved in PBS with 10% ethyl alcohol) was administered at 72 h and 48 h before the LPS challenge. reserpine (5 mg kg−1; i.p.; dissolved in 25% DMSO-PBS) was injected at 24 h before the lethal endotoxemia. fusaric acid (40 mg kg−1 per dose; i.p.) was administered at 8 h and 4 h before the LPS challenge. fenoldopam (10 mg kg−1 per dose; i.p.) was injected at 30 min and 5 min before LPS administration. pergolide (10 mg kg−1 per dose; i.p.; dissolved in 25% DMSO-PBS) was given at 12 h and 5 min before the LPS challenge. butaclamol (12 mg kg−1, i.p) was given 60 min before the endotoxemic challenge. In vitro; dopamine, fenoldopam, pergolide and butaclamol were used at 5 min before the LPS challenge at the concentration indicated in the figures.

Differentiation and Pharmacological Stimulation of LLCPK Cells

LLCPK cells plated on sterile glass-bottom plates were cultured to a confluent monolayer in Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10% fetal bovine serum at 37°C in 5% CO2. Serum was then withdrawn from cells to allow differentiation. A faster differentiation could be achieved by cell-cell contact inhibiting cell growth and/or withdrawing serum from the cell culture media. Because only fully-differentiated cells have well-developed cilia, cell differentiation can be validated by immunostaining of the sensory proteins in the cilia or responsiveness of the cells to fluid-shear stress. We have previously discussed cell differentiation in details (Nauli et al., 2013 (link)). Once differentiated, dopamine or fenoldopam (SigmaAldrich, Inc.) were gently added to culture plates to achieve 1 nM to 10 μM final concentrations for 16 h in the absence of serum. For control experiments, vehicle alone was added to cells in the same manner and volume as those contained pharmacological agents.

Fenoldopam Treatment in Sepsis Models

LPS (Escherichia coli 0111:B4), streptozotocin, glucose, dopamine hydrochloride, and fenoldopam were purchased from Sigma-Aldrich® (Saint Louis, MO, USA). Pentobarbital sodium was purchased from Diamondback (Scottsdale, AZ, USA); ketamine from Henry Schein animal health (Dublin, OH, USA); xylazine from Akorn animal health (Lake Forest, IL, USA), and enrofloxacin from Bayer Healthcare (Shawnee Mission, KS, USA). streptozotocin was injected (STZ; i.p., 50 mg/kg) at 10 and 5 days before the experiment as previously reported (35 (link), 36 (link)). Treatment with fenoldopam (Fen; 10 mg/kg/dose; i.p.) was administered at 6 and 1 h before LPS or CLP in most experiments. Treatment with fenoldopam was started 15 h after CLP and given every 12 h for 3 days in the survival experiments.
