Western Blot Analysis of Mevalonate Pathway Enzymes

Immunohistochemical Analysis of FDPS in Pancreatic Cancer

Tissue arrays comprised of normal pancreas (from donors for pancreas transplant) and PC patient samples were obtained from the Rapid Autopsy Program of the University of Nebraska Medical Center (PI: Michael Hollingsworth, IRB# 091-01-FB). The tissues were stained with anti-FDPS polyclonal antibody (ab38854, Abcam, Cambridge, MA, USA) using an antibody dilution of 1 : 100. The immunostaining technique was performed according to the standard laboratory procedure (Moniaux et al, 2008 (link)). The FDPS staining was scored by a pathologist using the criteria described in Supplementary Methods. Representative photographs were taken under a Nikon light microscope with a Q-capture Micropublisher 5.0 camera (Leeds Precision Instruments, Minneapolis, MN, USA) by utilising the Q-capture suite software package (QImaging, Surrey, BC, Canada).
