Clinical isolates of vancomycin-resistant E. faecium and E. faecalis (Table 1) were obtained from the Biodefense and Emerging Infections Research Resources Repository (BEI Resources) and American Type Culture Collection (ATCC). Brain heart infusion (BHI), Tryptic soy broth (TSB), Tryptic soy agar (TSA) and enterococcosel agar were purchased from Becton, Dickinson and Company (Cockeysville, MD, USA). Phosphate buffered saline (PBS) was purchased from Corning (Manassas, VA, USA).

Enterococcal Strains Protocol Acquisition

Production of Polyhydroxybutyrate in E. coli

For all experiments single colonies were picked from Luria–Bertani (LB) agar plates, inoculated in 5 mL LB media pre-cultures and grown overnight at 37 °C [29 ]. These 5 mL cultures were then used to start larger 50 mL cultures. Larger cultures consisted of a modified M9 minimal media containing: M9 salts (Na2HPO4, KH2PO4, NaCl, NH4Cl, Becton, Dickinson and Co, Sparks, MD), supplemented with 1.75 % (w/v) glucose (ACS grade, Acros Organics, Fair Lawn, NJ), 0.2 % (w/v) yeast extract (Becton, Dickinson and Co, Sparks, MD), and 100 µg/ml ampicillin (IBI Scientific, Peosta, IA). The addition of small amounts of yeast extract to the culture has been shown to increase PHB yields [27 (link), 30 (link)]. The 50 mL culture was grown overnight in an orbital shaker table at 37 °C and 220 rpm and was used to seed 1 L fermentors of the same media composition. For PHB production studies using fermentors, 0.1 mM Isopropyl β-d-1-thiogalactopyranoside (IPTG) (Gold Biotechnology, Inc. St. Louis, MO) was added at the start of the fermentation (t = 0 h).

Culturing and Enumerating Candida albicans

Candida albicans SC5314 was provided by Dr. Andrew Koh at the University of Texas Southwestern Medical Center [6 (link)]. This study used Yeast Peptone Dextrose (YPD) (242810, BD Difco, Franklin Lakes, NJ, USA) agar (BP1423, Fisher, Waltham, MA, USA), as well as Bacteroides Bile Esculin Agar (BBE) (AS114, Anaerobe Systems, Morgan Hill, CA, USA) for various assays. Additionally, the antibiotics kanamycin (J61272, Alfa Aesar, Tewsbury, MA, USA), ampicillin (69-52-3, IBI Scientific, Dubuque, IA, USA), and streptomycin (S6501, Sigma-Aldrich, St. Louis, MO, USA) were used in media for fungal enumeration, and cefoperazone (J65185, Alfa Aesar, Tewsbury, MA, USA) was used in antibiotic pre-treatment. Taurocholic acid (TCA) (16215, Cayman Chemicals, Ann Arbor, MI) and cyclophosphamide (PHR1404, Millipore Sigma, St. Louis, MO, USA) were purchased for these studies. Additional kits and reagents used included: hydrogen sulfide gas assay (ZAN-5084, Cell Biolabs, San Diego, CA), fluorescein isothiocyanate-dextran (FITC-dextran) (FD4, Millipore Sigma, St. Louis, MO, USA), and sodium formate (A17813, Alfa Aesar).
