Western Blot Analysis of CCR1 in PBMCs

Fresh PBMC were washed twice with cold PBS and lyzed with lysis buffer (50 mm Tris–Cl, pH 7·4, 0·15 m NaCl, 1 mm EDTA, 1% Triton X‐100, 10 μg/ml each of aprotinin and leupeptin, and 1 mm phenylmethylsulphonyl fluoride). The protein (30 μg) lysate was electrophoresed on a 10% sodium dodecyl sulfate–polyacrylamide gel. The proteins were transferred onto nitrocellulose membranes (Protran; GE Healthcare Life Sciences, Chicago, IL, USA) and subjected to immunoblot analysis using antibodies. The immunoblots were developed with the enhanced chemiluminescence system (GE Healthcare Life Sciences). Antibody against CCR1 (MAB5986">MAB5986) was purchased from R&D Systems and glyceraldehyde 3‐phosphate dehydrogenase (GAPDH) antibody (sc‐32233) was from Santa Cruz Biotechnology, Inc. (Dallas, TX).
