Monomeric sirna

T7 promoter primer, amino-cholesteryl (5′- and 3′-)-conjugated DNA, 3′-Cy5 labeled DNA, monomeric siRNA, and primers for qRT-PCR were all obtained from Bioneer Co. (Korea), as well as 5′-phosphorylated ssDNA templates for RCT reaction of various RNA nanoparticles including Dsi RNPs. The branched PEI (25 kDa) polymer was purchased from Sigma-Aldrich (UK). All oligonucleotide sequences are listed in Table S1.

Linear ssDNA template, T7 promoter primer, 3′-cholesterol-modified DNA conjugate (DNA–Chol), 5′-amine-modified DNA fragment for preparation of FA–DNA, 3′-Cy5-labelled and 5′-amine-modified DNA fragment for preparation of FA–DNA-Cy5, primers for qRT–PCR, scrambled sense siRNA, scrambled antisense siRNA and monomeric siRNA were all synthesized from Bioneer Co. (Daejeon, Korea). All DNA sequences are summarized in Supplementary Table 1. 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimide (EDC), Sulfo-NHS and folate were purchased from Sigma (St Louis, MO, USA). Lipofectamine2000 was purchased from Invitrogen (USA). Cell culture media were obtained from WelGENE (Korea). The RNA mfold program (available in http://mfold.rna.albany.edu/?q=mfold/RNA-Folding-Form) was used to predict the RNA secondary structures45 (link). Female BALB/c nude mice and female C57BL/6J mice were purchased from Orient Bio Inc. (Korea).
