GV Oocyte Maturation Inhibition Assay

GV oocytes from wild-type mice were cultured in M16 medium containing IBMX (both Sigma) at 37°C, 6% CO2 and 5% O2, and supplemented with: BIX-10294 (Stratech) at final concentrations of 0.1 µM and 1 µM (Huang et al., 2017 (link)); MM-102 (Cayman Chemicals) at final concentrations of 6 µM or 60 µM, within the range previously reported (Zhang et al., 2018 (link)); or a combination of 1 µM BIX-01294 and 60 µM MM-102. The inhibitors were dissolved in DMSO (Sigma) and an equivalent concentration was used for the control groups. After 48 h the surviving GV oocytes were either fixed in 4% paraformaldehyde (Thermo Scientific) for investigation by IF, or frozen at −80°C for RNA isolation and subsequent qRT-PCR.
