Nitric Oxide Production Assay

RAW264.7 cells were seeded in triplicate in 96-well plates at a density of 5 × 104 cells/well and incubated under standard conditions for 24 h. The cells were treated with 20 ng/mL of murine recombinant IFNγ (GIBCO, Gaithersburg, MD, USA), with or without (control) compounds at various concentrations for 24 h. The level of NO production was evaluated by determining the nitrite concentration in culture medium with a Griess Reagent System (Promega, Madison, WI, USA). The IC50NO value was calculated as the derivative concentration required to decrease NO production to 50% of the non-treated control.
