Quantifying Mg Ion Levels in Soil Samples

MgCl2 was purchased from Daejung (Seongnam, Korea). Two M of stock solution was prepared by dissolving 4.1825 g of MgCl2 hexahydrate in 10 mL of deionized water. The filter-sterilized stock solution with Millex® syringe filter was then serially diluted with deionized water. The final concentrations of diluted solution were 0.0, 0.004, 0.02, 0.04, 0.2, 0.4, 0.8, 1.2, 1.6, and 2.0 M. One hundred μL of each reagent was spiked into 1 g of sand sample. This means the sand samples would contain 0.001, 0.005, 0.01, 0.05, 0.1, 0.2, 0.3, 0.4, and 0.5% (w/w) Mg ion content, respectively.
Subsequently, three inhibitors laden sands were subject to both methods with and without purification. Five μL of the lysates or gDNA (triplicate sample) were then used for subsequent qPCR assay.

Hygroscopic Equilibrium of UPS Powder

Eight different salts were chosen, including LiCl, KCH3CO2, MgCl2, K2CO3, Mg(NO3)2, KI, (NH4)2SO4, and KNO3 (Daejung Chemical & Metals Co., Ltd, Seoul, Korea), to have 11%, 23%, 33%, 43%, 53%, 69%, 81%, and 93% RH, respectively. The experimental setup comprised 64 desiccators (8 × 8 batches, each containing 25 g UPS powder). The 25 g samples were uniformly dispersed on five sterile aluminum Petri plates and transferred to disinfected and sterile desiccators equipped with stands with several 5 mm holes in diameter holding 850 mL saturated salt solution. After being relocated to a chamber with constant temperature control of 25 °C, the desiccators were hermetically sealed. The RH within the desiccators and the MC of the samples were measured at intervals until hygroscopic equilibrium was reached. After 1 week of keeping hygroscopic equilibrium, the 4 week storage period began. After 2, 3, and 4 weeks, the samples were randomly obtained from each desiccator for analysis.
