Proteomics Analysis of Extracellular Vesicles

The culture medium was Multicell RPMI 1640, 1× obtained from Wisent Inc.; Saint-Jean-Baptiste, QC, Canada (350-005-CL). Nalgene 0.45 μm pore PES rapid flow vacuum filter units (165-0045) and Pierce silver stain kit (24612) were from ThermoFisher; Waltham, United States. Satorius vivacell 100 PES concentrators were purchased from Fisher Scientific (VC1042).

Characterization of Liver Spheroid Model

Liver spheroids and tissues were fixed in neutral buffered formalin for 1 day at room temperature and submitted to HistoWiz Inc. (Brooklyn, NY, USA) for paraffin processing and sectioning. Paraffin sections (4 µm) were analyzed by direct fluorescence microscopy, H&E staining and IHC. IHC antibodies included anti-HNF4α (PP-K9218-00, R&D Systems, Minneapolis, MN, USA; 1:125); anti-αSMA (ab124964, Abcam; 1:1000); anti-Ki67 (ab16667, Abcam, Cambridge, MA, USA; 1:200); anti-CK19 (ab133496, Abcam; 1:250); anti-CD68 (ab125212, Abcam; 1:1000); anti-CD31 (ab12443, Abcam; 1:1000), anti-albumin (ab192603, Abcam; 1:50) and anti-HSEC (NB110-68095, Novus Biologicals, Centennial, CO, USA; 1:250). For the co-spheroids, direct ZsG and tdT fluorescence images were taken from sections prior to IHC. HNF4A+ and Ki67+ counting was performed on three spheroids each from three independent batches of spheroids and plotted using GraphPad Prism 9.4.

Immunohistochemical Analysis of RGCC Expression in Colorectal Cancer

The tumor and its paired
adjacent tissues of colorectal cancer patients were fixed with 4%
paraformaldehyde, embedded in paraffin, dewaxed, hydrated, and antigen-repaired.
After being blocked by 5% BSA (AB192603, Abcam), the tissues were
incubated by the RGCC antibody (BS-9079R, 1:200, Bioss Inc, China)
overnight at 4 °C. After being washed with phosphate-buffered
saline (PBS), the samples were incubated with the horseradish peroxidase
(HRP)-labeled broad-spectrum secondary antibody (D-3004, Shanghai
Changdao Biotechnology Co., Ltd., Shanghai, China) at 37 °C for
30 min. After dyeing with DAB (FL-6001, Shanghai Changdao Biotechnology
Co., Ltd., Shanghai, China) and hematoxylin, a Leica microscope (DM1000)
was used to collect photographs. The mean optical density (IOD) of
RGCC expression in each sample was calculated by Image-Pro Plus 6.0
analysis software. All experimental procedures were carried out in
accordance with Chinese legislation regarding medical ethics and were
approved by the Medical Ethics Committee of Zunyi Medical University
((2019) H-008).

Histological Analysis of Mouse Liver Tissues

Mouse liver tissues were fixed in formalin for 24 h and embedded in paraffin. Five-micron-thick sections were made from paraffin-embedded liver tissues and then stained with haematoxylin and eosin to detect histological structure and with Sirius red to detect liver fibrosis. For immunohistochemical staining, the sections were incubated with primary antibodies to ALB (ab192603, 1:300, Abcam) and CD68 (ab955, 1:200, Abcam) according to previously described protocols [16 (link)].

Functionalized POZ Polymers for Biomedical Applications

Biosol was purchased from National Diagnostics. Pharmaceutical grade human serum albumin (HSA), used in the clinical grade trileaflet BHV experiments, was purchased from Octapharma. THP-1 cells, a monocyte/macrophage cell line, were purchased from ATCC. RPMI medium was purchased from Cell Culture Technologies. TNF-α ELISA was obtained from Invitrogen. Cellulose dialysis membrane was purchased from Spectrum Labs (10768-700). The Von Kossa staining kit was obtained from Abcam (ab150687). All chemicals were purchased from Sigma Aldrich, unless otherwise stated.

Histopathological and Immunohistochemical Analysis of Liver and Kidney Tissues

4-µm-thick formalin-fixed paraffin-embedded liver and kidney tissue sections were used for histopathology and immunohistochemistry analyses. Comprehensive Histological Analysis of Liver Samples
