adjacent tissues of colorectal cancer patients were fixed with 4%
paraformaldehyde, embedded in paraffin, dewaxed, hydrated, and antigen-repaired.
After being blocked by 5% BSA (AB192603, Abcam), the tissues were
incubated by the RGCC antibody (BS-9079R, 1:200, Bioss Inc, China)
overnight at 4 °C. After being washed with phosphate-buffered
saline (PBS), the samples were incubated with the horseradish peroxidase
(HRP)-labeled broad-spectrum secondary antibody (D-3004, Shanghai
Changdao Biotechnology Co., Ltd., Shanghai, China) at 37 °C for
30 min. After dyeing with DAB (FL-6001, Shanghai Changdao Biotechnology
Co., Ltd., Shanghai, China) and hematoxylin, a Leica microscope (DM1000)
was used to collect photographs. The mean optical density (IOD) of
RGCC expression in each sample was calculated by Image-Pro Plus 6.0
analysis software. All experimental procedures were carried out in
accordance with Chinese legislation regarding medical ethics and were
approved by the Medical Ethics Committee of Zunyi Medical University
((2019) H-008).
