Quantitative Papain Activity Assay

Sodium acetate (anhydrous, ≥99%), trichloroacetic acid (≥99%), and 2,4,6-Tris(2-pyridyl)-s-triazine (≥98%) were obtained from the Sigma-Aldrich group (Schnelldorf, Germany). Citric acid (99%), potassium sulfate (≥99%), copper sulfate (≥99%) sulfuric acid (≥99%), phenolphthalein (≥98%), methanol (≥98%), glucose (≥99%), phenol (≥99%), and sodium hydroxide (≥99%) were purchased from Reanal (Budapest, Hungary). Ferric chloride (≥99%), sulfuric acid (≥99.9%), amyl alcohol (≥99.9%), ascorbic acid (99.7%), bacteriological agar powder, and soybean casein digestive medium were procured from Merck (Darmstadt, Germany). Acrylamide (≥99%), sodium-dodecyl sulfate (≥99%), ammonium persulfate (≥99%), Tetramethylethylenediamine (≥99%), tris(hydroxymethyl)aminomethane (≥99%), glycine (≥99%), ethyl alcohol (≥99%), coomassie blue stain (≥99%), acetic acid (≥99%), isopropanol (≥99%), glycerol (≥99%), 2-βmercaptoethanol (≥99%), and bromophenol blue (≥99%) were purchased from BIO-RAD (BIO-RAD, USA). Milli-Q ultrapure deionized water (18.2 MΩ·cm; Merck-Millipore, Molsheim, France) was used throughout the experiment.

The following reference compounds were purchased from Merck Group (Shanghai, China): dichloromethane (99.8%), N,N-dimethylmethanamine (p.a.), and chloroform (p.a.) 2-methyl-1-propanal, diacetyl, hexanal, 2-pentyl furan, amyl alcohol, 2-methyl pyrazine, octanal, acetone alcohol, 2,6-dimethyl pyrazine, 2-ethyl pyrazine, 2,3-dimethyl pyrazine, dimethy trisulfide, nonanal, 2-ethyl-6-methyl pyrazine, 2,3,5-trimethyl pyrazine, trans-2-octenal, 1-octen-3-one, 2,3-dimethyl-5-ethylpyrazine, 2,3,5,6-tetramethyl pyrazine, benzaldehyde, (E)-2-nonenal, 2-propyl-pyridine, gamma-butyrolactone, furan-2-ylmethanol, 2-hexylthiophene, 2-undecenal, (E,E)-2,4-decadienal, and (R)-(-)-pantolactone. The alkane mixes C5-C40 were purchased from Anpel (Shanghai, China). The purity of these and the aforementioned compounds was not less than 97%.

Colorimetric Indole Quantification Assay

The Kovacs assay protocol was modified from the method of Darkoh et al. [34 (link)] in order to detect indole in the range of 10–40 μM as well as the normal sensitivity range of 50–500 μM. 200 μL of culture was centrifuged at 11,337 x g (Eppendorf, Minispin Plus, Hamburg, Germany) for 2 min. 100 μL of the supernatant was mixed with 150 μL of Kovacs reagent (5 g p-dimetylaminobenzaldehyde, Sigma, Belgium; 75 mL amyl alcohol, Sigma, Germany; 25 mL 37% w/w HCl, Fisher Scientific, Loughborough, UK) and incubated at room temperature for 5 min. The absorbance of the reaction mix was measured at 530 nm using a SpectraMax 190 microplate reader (Molecular Devices, Sunnyvale, USA). The concentration of indole was calculated using a standard curve generated with indole (Sigma, USA) standards dissolved in ethanol.

Phenolic Compounds Extraction and Analysis

Whey Bioconversion by Yeast and Enzymes

Whey produced during Swiss cheese production was obtained from the Key Laboratory of Dairy Science, Northeast Agricultural University, Harbin, China, and all whey samples were stored at -20°C until processing. Sweet whey was made using liquid bovine rennet (containing 95% chymosin and 5% pepsin) obtained from New England Cheesemaking Supply Company, as earlier described by Timón et al. (2014) (link). Lactase (10,000 acid lactase unit/g, 37°C, pH 6) was bought from the Nanning Pangbo Bioengineering Co. Ltd. We purchased α-dlactose monohydrate, d-galactose, and d-(+)-glucose compounds from Shanghai Gold Wheat Biological Technology Company, prepared from commercially available standards, purity grade ≥98%. Ethyl acetate, acetaldehyde, acetic acid, methanol, n-butanol, 1-propanol, 2-butanol, amyl alcohol, isobutyl alcohol, isoamyl alcohol, and 4-nanonol (GC standard purity) were purchased from Sigma-Aldrich as internal standards.
