Culturing Cancer and Stem Cells

Transfection of Thyroid Cancer Cells

Human thyroid cancer SW579 and KTC-1 cells and human normal thyroid epithelial cells Nthy-ori 3-1 were bought from Cell Bank of Chinese Academy of Sciences (Shanghai). Cells were cultivated in a sterile incubating device under 37°C, 5% CO2, and 5% v/v. TREM2 siRNA and its negative control (NC) were constructed by GenePharma (Shanghai, PRC). Blank plasmids were used as controls. SW579 cells at logarithm growth phase were inoculated in 6-well dishes and transfected with control, NC, and TREM2 siRNAs at 70%–80% cell fusion after following the instruction manual of Lipofectamine 2000 (Invitrogen; Thermo Fisher Scientific, Inc.). After 48 h of transfection, total RNA was obtained, and RT-PCR was conducted to demonstrate the efficiency of transfection.

Thyroid Cancer Cell Lines and Tissues

Human thyroid cancer cell lines CAL-62, KHM-5M, BHT-101, B-CPAP, and normal thyroid cell lines Nthy-ori-3-1 were purchased from the China Center for Type Culture Collection (CCTCC, China). A total 15 of DTC patients who underwent radical thyroidectomy in Ruijin Hospital of Shanghai Jiaotong University Medical College and were confirmed as DTC by postoperative histopathological examination were included in this study. All samples were obtained with the patients’ informed consent, and the samples were histologically confirmed by at least 2 pathologists independently in a double-blinded fashion.
