Immunophenotypic Analysis of AML Cell Lines

For immunophenotypic analysis AML cell lines were plated at 4 × 105 cells/mL into 6 well plates and, the day after, 1 × 106 cells were collected and stained with the following fluorochrome-conjugated anti-human antibodies: CXCR4 (PE, R&D Systems) and CD18 (ITGB2, APC, BD Pharmingen). Staining was performed at 4°C for 20 minutes in the dark.
1 × 105 events were acquired at flow cytometer (FACS Canto II, Becton Dickinson). Surface expression (percentage of positive cells) and MFI (mean fluorescent intensity) values were calculated for CXCR4 and ITGB2 with FlowJo software.
