For determination of IL-2 and IFN-γ production by PBMC, cells were activated and treated with C. ipecacuanha fractions, cyclic peptides, or CsA for 48 hrs. PBMC were then re-stimulated using phorbol-12-myristate-13-acetate (PMA; 50 ng/mL) and ionomycin (500 ng/mL; both from Sigma) and incubated with BD Golgi Plug™ (BD Biosciences) for 4 hrs. After washing, cells were fixed with 4% paraformaldehyde (PFA; Morphisto) and staining of intracellular cytokines was carried out using PE-conjugated anti-human IL-2 or PE-conjugated anti-human IFN-γ mAbs (both from Affymetrix). Levels of cytokine production were determined using flow cytometric analysis of three independent experiments.
