Vigantol 20.000 IU/mL was obtained from Merck Serono (Mumbai, India). Vitamin D deficient (EF R/M, E15312-24) and conventional rat chow (SM R/M, S8106-S011) were purchased from ssniff Spezialdiäten GmbH (Soest, Germany). Pentobarbital was obtained from Ceva-Phylaxia (Budapest, Hungary). The composition of normal Krebs-Ringer solution (in mmol/L) was: NaCl 119; KCl 4.7; NaH2PO4 1.2; MgSO4 1.17; NaHCO3 24; CaCl2 2.5; glucose 5.5 and EDTA 0.034. The composition of Ca2+-free Krebs solution (in mmol/L) was: NaCl 92; KCl 4.7; NaH2PO4 1.18; MgCl2 20; MgSO4 1.17; NaHCO3 24; glucose 5.5; EGTA 2 and EDTA 0.025. Uridine 5’-triphosphate (UTP) and bradykinin were purchased from Sigma-Aldrich (Darmstadt, Germany). Anti-eNOS [14 (link)] and anti-COX-2 [14 (link)] antibodies were obtained from Abcam (Cambridge, MA, USA). Anti-smooth muscle actin antibody (1A4) [15 ] and UltraView Universal DAB Detection Kit were obtained from Ventana Medical Systems, Inc. (Tucson, AZ, USA). The C Kinase Activity Reporter (CKAR) was previously described28 (link). mCherry-tagged constructs were cloned into pcDNA3 with mCherry at the N-terminus at the BamHI and XbaI sites. All mutants were generated by QuikChange II Site-Directed Mutagenesis (Agilent). Phorbol 12,13-dibutyrate (PDBu) and uridine-5’-triphosphate (UTP) were purchased from Sigma-Aldrich.

PLK4 Activation Loop Phosphorylation Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
The pan anti-phospho-PKC activation loop antibody was described previously 107 (link). The anti-phospho-PKCα/βII (T638/641; 9375S) and pan anti-phospho-PKC (βII S660; 9371S) antibodies were purchased from Cell Signaling. Anti-PKCβ antibody was purchased from BD Transduction Laboratories. The anti-α-tubulin (T6074) antibody was from Sigma. Phorbol 12,13-dibutyrate (PDBu) and Uridine-5-triphosphate (UTP) were purchased from Sigma-Aldrich. 6His-N-terminally tagged human PLK4 (amino acids 1-269) was expressed and purified as described previously 108 (link) and affinity purified using Ni-NTA agarose. Proteins were eluted from beads by incubation with 0.5 M imidazole. Described PLK4 mutations were introduced using standard PCR-based site-directed mutagenesis protocols and confirmed by sequencing the PLK4 1-269 coding region. pT170 PLK4 phosphospecific antibody was raised in rabbits against a phosphorylated PLK4 T-loop consensus peptide, and purified using standard affinity protocols prior to storage at -20°C 59 (link). The PLK4 inhibitors VX680 and staurosporine and the PLK1 inhibitor BI2536 were employed after dilution from 50 mM DMSO stocks stored at -80°C 79 (link).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
