Isolation and Characterization of Murine Hematopoietic Stem Cells

Purification of Spermatogonia via FACS

Fluorescence-activated cell sorting (FACS) purification was performed on cultured cells to remove contaminating feeder cells prior to RNA extraction or subsequent analyses or western blot. Following culture and collection as described above, samples were washed in 2% v/v FBS in PBS and stained with phycoerythrin (PE)-conjugated EpCAM (1:300, eBioscience, clone G8.8) for 30 min at 4 °C prior to washing and resuspension in 2% v/v FBS in PBS. DAPI was added to final cell suspensions for live/dead cell discrimination. Spermatogonia were purified according to their expression of EpCAM10 (link). For cultured cells expressing constructs tagged with GFP or tdTomato, spermatogonia were isolated based on endogenous fluorescence of these tags, without staining for EpCAM. Isolation of spermatogonia from testes of control and Plzf–/– mice was performed following staining with allophycocyanin (APC)-conjugated anti-cKIT (Biolegend, clone 2B8, #105812), PE-conjugated anti-CD49f (Biolegend, clone GoH3, #313612), PE-Cyanin7-conjugated anti-EpCAM (Biolegend, clone G8.8, #118216) and fluorescein isothiocyanate (FITC)-conjugated anti-CD9 (eBioscience, clone KMC8, #11-0091-82)12 (link). FACS was performed at Monash University FlowCore using a BD Influx Cell Sorter (BD Biosciences).

Antibody-based Immunoassay Protocol

Multiparameter Flow Cytometry of Hematopoietic Cells

The whole blood was stained with APC-conjugated anti-Ter119 and PE-conjugated anti-CD61 for erythrocyte and platelet analysis. White blood cells after RBC lyses were stained with PE-conjugated anti-CD8, PE-Cy7–conjugated anti-B220, APC-conjugated anti–Mac-1, APC-Cy7–conjugated anti-CD4, and Pacific blue (PB)–conjugated anti-Gr1 (BioLegend). Single cell suspensions from BM and spleen were stained with lineage-marker cocktail followed by FITC-conjugated anti-CD41, PE-conjugated anti-CD150, PE-Cy7–conjugated anti-CD48, APC-conjugated anti–c-Kit, BV421-conjugated anti–Sca-1, and streptavidin-APC-Cy7. For progenitor analysis, cells were stained with lineage-marker cocktail followed by FITC-conjugated anti-CD34 (BD), PE-conjugated anti-CD16/32, APC-conjugated anti–c-Kit, BV421-conjugated anti–Sca-1, and streptavidin-APC-Cy7 antibodies (BioLegend). For megakaryocytes and erythrocyte precursor analysis, cells were stained with FITC-conjugated anti-CD42c (Emfret), Dylight-647–conjugated anti-αIIbβ3 (CD41/61; Emfret), PE-conjugated anti-CD71, and APC-conjugated anti-Ter119. Analyses were performed on an LSRFortessa (BD) or CyAn ADP Analyzer (Beckman Coulter), and results were analyzed with FlowJo software.

The purified anti-mouse TER-119, Gr-1, B220, CD3 and PCNA and PE-conjugated anti-mouse TER-119 antibodies were purchased from BD Pharmingen (San Diego, CA, USA). APC-conjugated CD71, PE-conjugated anti-Sca1 and APC-conjugated anti-c-kit antibodies were purchased from Biolegend (San Diego, CA, USA). Biotinylated anti-c-kit antibody was purchased from Abcam (Cambridge, UK). Normal rabbit IgG, biotinylated goat anti-rat IgG, biotinylated goat anti-mouse IgG, biotinylated goat anti-hamster IgG, Texas Red-conjugated goat anti-Rat IgG antibodies and an avidin-biotin complex kit (ABC Elite standard kit) were purchased from Vector Laboratories (Burlingame, CA, USA). The anti-mouse F4/80 antibody was obtained from AbD Serotec (Kidington, UK). The Lineage Cell Depletion kit was purchased from Miltenyi Biotec (Bergisch Gladbach, Germany).
