Quantification of Amlodipine in Microencapsulated Probiotic

Microencapsulated L. plantarum IS-10506, which is a novel probiotic that is isolated from dadih (a traditional Indonesian fermented buffalo milk), was prepared as previously described.10 (link) Amlodipine (amlodipine besylate), the internal standard nortriptyline hydrochloride (NOR), and 4-chloro-7-nitrobenzofurazan (NBD-Cl) were purchased from Sigma Aldrich (Singapore). Methanol, acetonitrile, ethanol, sulfuric acid, hydrochloric acid, boric acid, and sodium hydroxide were purchased from Merck (Darmstadt, Germany). All reagents that were used were analytical grade, except for Methanol, which was high-performance liquid chromatography (HPLC) grade.
Stock solution of amlodipine was prepared by dissolving 10 mg of free base with 2 mL ethanol and diluting to 100 mL with water. A plasma standard with serial dilution of 2.5, 10, 40, 160, 640, and 1280 ng/mL was prepared to provide plasma calibration samples. NOR was prepared at a concentration of 100 µg/mL and diluted to obtain 50 ng/mL of working solution. NBD-Cl solution (0.08%, w/v) was freshly prepared in Methanol. Teorell and Stenhagen buffer solution13 was composed of phosphoric acid, citric acid, 0.1 M sodium hydroxide, and 0.1 M hydrochloric acid, and adjusted to a pH of 8.6.

Keck Biotechnology Resource Laboratory (New Haven, CT), respectively.

Phenolic Standards Characterization

Nine phenolic standards including gallic acid (GA), syringic acid (SRA), p-coumaric acid (pCA), ferulic acid (FA), sinapic acid (SNA), cinnamic acid (CNA), protocatechuic acid (PRA), rutin (RUT) and quercetin (Q), 2,2-diphenyl-1-picrylhydrazyl (DPPH reagent), 2,2-azinobis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt (ABTS reagent), 4-chloro-7-nitrobenzofurazan (NBD-Cl), ammonium acetate, and neocuproine were purchased from Sigma-Aldrich (St. Louis, MO, USA). In all cases, the purity of the standards exceeded 98%. Aluminum chloride (AlCl3) was obtained from AcrossOrganics (Morris Plains, NJ, USA) and high-performance liquid chromatography (HPLC)-grade acetonitrile (ACN) was obtained from Avantor (Central Valley, PA, USA) while the other reagents were obtained from POCh (Gliwice, Poland). The redistilled water was prepared by triple distillation of water in a Destmat® Bi-18 system (Heraeus Quarzglas, Hanau, Germany).

Synthesis and Functionalization of GA-AgNPs

Salmon sperm dsDNA was purchased from Sigma Aldrich (Slovakia) and its 0.1 mg mL−1 stock solution prepared in PB was stored at 4 °C. 4-Chloro-7-nitrobenzofurazan (NBD-Cl) was purchased from Sigma Aldrich (Slovakia) and prepared as a stock solution of 2 mg mL−1 (10 mmol L−1) in acetonitrile. GA-AgNPs were synthesized in our laboratory using the protocol described below. Silver nitrate, gallic acid, sodium hydroxide, ammonia, sodium hydrogen phosphate, sodium dihydrogen phosphate, and other required chemicals were obtained from Mikrochem (Slovakia). For GA-AgNPs modification, doxorubicin (DOX) was purchased from Sigma Aldrich (Slovakia) and its 1 mg mL−1 stock solution was prepared in deionized water and kept in dark.

Analytical Method for TPM Quantification

TPM was obtained from Sequoia Research Products (Pangbourne, UK), while bendroflumethiazide as an internal standard (IS) and 4-Chloro-7-nitrobenzofurazan (NBD-Cl) as a derivatization reagent were obtained from Sigma Aldrich (Steinheim, Germany). HPLC grade methanol and acetonitrile were purchased from Sigma Aldrich (Steinheim, Germany). All other chemicals used, including dichloromethane, diethyl ether, ethyl acetate, hexane, boric acid, potassium dihydrogen phosphate, sodium hydroxide and formic acid all from Merck (Darmstadt, Germany) were at least analytical grade. Water for all applications was obtained from a Q-POD Milli-Q water purification system (Millipore Corp., Billerica, USA) with resistivity ≥18.2 MΩcm.
