Radiolysis of Lipid Liposomes

HOCH 2 CH 2 SH and 1-palmitoyl-2-oleyl-sn-glycero-3-phosphocholine (POPC) were purchased from Aldrich. 1-Palmitoyl-2-linoleoyl-sn-glycero-3-phosphocholine (PLPC) and 1-stearoyl-2-arachidonoyl-snglycero-3-phosphocholine (SAPC) were purchased from Avanti Polar Lipids (Alabaster, AL) (see Fig 1B). All solvents were HPLC-grade and the utilized water was glass-distilled. Radiolysis was performed at room temperature (22 ± 2 °C) on 1 mL samples using a 60 Co Gammacell. For the construction of 100 nm liposomes the Mini Extruder was used from Avanti Polar Lipids. The composition of lipids during the experiments was monitored by gas chromatography using the GC 7890Β from Agilent equipped with a flame ionization detector. The column used was the DB-23 capillary column (60m × 0.25mm). The heating was carried out at a temperature of 165 °C for 3 minutes followed by an increase of 1 o C/min up to 195 °C. The temperature remained at 195 °C for 40 minutes and then it was followed by an increase of 10 °C /min up to 240 °C for 10 minutes.
