Characterization of Pharmaceutical-Grade MA Formulation

MA (purity > 99.5%) was received as a kind gift sample from Orchid Pharmaceutical Ltd. (Roorkee, India). Hydroxypropyl methylcellulose-K4M (HPMC-K4M), sodium dodecyl sulfate (SLS), polysorbate 80 (Tween ® 80), hydrochloric acid (HCl), methanol (MeOH), dimethyl sulfoxide (DMSO), sodium hydroxide (NaOH), potassium dihydrogen phosphate (KH 2 PO 4 ), sodium chloride (NaCl), ethanol (C 2 H 5 OH), potassium chloride (KCl), disodium hydrogen phosphate dodecahydrate (Na 2 HPO 4 .12H 2 O), N,N-dimethylformamide (DMF), and acetonitrile (CH 3 CN) were commercially obtained from Central Drug House Ltd. (New Delhi, India). The water utilized for all experiments was high-purity/deionized water, which was obtained using a Milli-Q ® water purification unit (Milli-Q Co., Lyon, France). All other materials used were of analytical/pharmaceutical grade.

Silicone and Paraffin Oil Nanoemulsion

Silicone, paraffin oil, and surfactant (Tween 80) were procured from Central Drug House (New Delhi, India). Silicone oil is a nonpolar solute and has efficient gaseous absorption capacity which makes it suitable oil for our experiment. Paraffin oil is a mixture of C10–C15 alkanes and cycloalkanes which are colorless, antioxidative, and have low viscosity.
The nonionic surfactant Tween 80 (Polyoxyethylene (20) sorbitan monooleate) with the hydrophilic–lipophilic balance of 18, was used for the formulation of silicone and paraffin oil nanoemulsion. The surfactant action is to reduce the interfacial tension between two phases [26 ].

Investigating Anxiolytic Mechanisms of Action

Diazepam (DZP, Svizzera Healthcare, India) and Buspirone HCl (BSP, Sigma-Aldrich, U.S.A) was used as reference drugs. Flumazenil (FLU, Sigma-Aldrich, U.S.A), a competitive inhibitor of GABAA receptor in the benzodiazepine site and WAY-100635 maleate salt (WAY, Sigma-Aldrich, U.S.A) a 5-HT1A receptor antagonist was used to find out the possible mechanism(s) of action. PNEO was dissolved in 0.01% (v/v) Tween 80 (Central Drug House, India) in saline and treated orally (p.o.). For the intraperitoneal treatments, the reference drugs and other agents were given by dissolving in normal saline (0.9 %w/v NaCl). All samples were freshly prepared before the behavioral testing and treated at 0.1 ml/10 g of body weight (b/w), except Flumazenil, which was treated at 0.2 ml/10 g of b/w.
