Extraction and Characterization of Acridones

Ten acridones were extracted from the stems of Atalantia monophylla, including N-methylatalaphylline, atalaphylline, N-methylatalaphyllinine, atalaphyllinine, N-methylcycloatalaphylline A, citrusinine I, citrusinine II, glycosparvarine, citruscridone, and buxifoliadine C (Figure 1). The isolation and structural elucidation is described elsewhere [45 (link)]. Before use, all acridones were prepared as stock solution in ethanol or DMSO at a concentration of 10 mM. Acetylthiocholine iodide (ATCI), beta amyloid 1-42 (Aβ1-42), tacrine, trolox, trypsin, fetal bovine serum (FBS), and Dulbecco’s modified Eagle medium nutrient mixture F-12 (DMEM/F12) were purchased from Sigma-Aldrich (SM Chemical supplies Co., Ltd., Bangkok, Thailand), Merck (Merck, Bangkok, Thailand), Gibthai (GT Chemical supplies Co., Ltd., Bangkok, Thailand), and Fluka (SM Chemical supplies Co., Ltd., Bangkok, Thailand). Human neuroblastoma cell line SH-SY5Y cells were purchased from ATCC (A.N.H. Scientific Marketing Co., Ltd., Bangkok, Thailand). AutoDock program version 4.2.6 (the Scripps Research Institute, La Jolla, CA, USA) and BIOVIA Discovery Studio 2017 (BIOVIA, San Diego, CA, USA) were used in the molecular docking study.

Isolation and Characterization of Coumarins from Toddalia asiatica

Multicomponent Herbal Blend for Neuroprotection

Isolation and Characterization of Human Cells

Trypsin, collagenase II, fetal bovine serum (FBS), dispase II, phosphate buffer solution (PBS, pH 7.2), penicillin (10,000 IU/mL), and streptomycin (10 mg/mL), were obtained from Invitrogen (Waltham, MA). Dulbecco's Modified Eagle Medium/Nutrient Mixture/F‐12 (DMEM/F‐12), Trypan Blue, p‐formaldehyde, poly‐L‐lysine, Triton X‐100, dimethyl sulfoxide (DMSO), propidium iodide (PI), and L‐glutamine were purchased from Sigma‐Aldrich (Saint Louis, MO). Ethylenediaminetetraacetic acid (EDTA) was obtained from Promega (Madison, WI). Purified antibodies, including anti‐neutrophil elastase (NE), anti−α‐SMA, and anti‐human nuclear antigen (HNA), were purchased from Abcam (Cambridge, England). Alexa Fluor 488‐ and Alexa Fluor 594‐conjugated goat anti‐rabbit antibodies and a Qtracker 655 cell‐labeling kit were obtained from LifeTechnologies (Eugene, OR). Flourescein isothiocyanate (FITC)‐conjugated goat anti‐mouse antibody was obtained from Santa Cruz Biotechnology (Dallas, TX). Purified antibody Ki‐67 was purchased from Cell Marque (Rockline, CA). Bovine serum albumin (BSA) was obtained from Calbiochem (San Diego, CA). T75 cell culture flasks and 24‐well plastic plates were purchased from Corning Inc. (Corning, NY). Vectashield mounting medium was purchased from (Vector Laboratories (San Diego, CA). Polymorphoprep was obtained from Axis‐Shield (Oslo, Norway).

Ovarian Cancer Cell Line Cultivation

In this study, two ovarian cancer cell lines, namely OVCAR-3 and SK-OV-3, were used. OVCAR-3 cells, isolated in 1982 from the malignant ascites of a patient with progressive adenocarcinoma of the ovary, were obtained from American Type Culture Collection (ATCC, Manassas, VA, USA). SK-OV-3 is an epithelial morphology cell line derived from the ovary of a 64-year-old, White female with ovarian adenocarcinoma. Both were maintained in the laboratory using Dulbecco’s Modified Eagle Medium/Nutrient mixture F-12 (DMEM/F12) (Sigma-Aldrich, Munich, Germany), supplemented with 20% Fetal Bovine Serum (FBS), and 1% penicillin-streptomycin (PS) (Sigma Chemical Co., St. Louis, MO, USA). Cells were incubated at 37°C in a humidified atmosphere containing 5% CO2.
