Bone marrow from femurs and tibias of ~8–12 week old mice were cultured in RPMI (BioWhittaker, MD) with 10% fetal bovine serum (HyClone Laboratories, Utah), 0.005% 2-mercaptoethanol,10 mg/L gentamicin (MP Biomedicals, LLC) and 10 ng/ml GM-CSF (Peprotech) for 6 days at 37°C and 5% CO2.
