Comprehensive Mouse Biosampling Protocol

Before blood sample collection, the body weight of each mouse was weighed and recorded with electronic balance (Shanghai Sunny Hengping Scientific Instrument, China). Full blood samples of mice were collected by extirpating eyeballs. Automatic blood coagulation analyzer (C2000, Perlong Medical, China) was used for the blood coagulation examination. Automatic hematology analyzer (Sysmex, Kobe, Japan) was used for blood routine examination. After sacrifice, the organs of mice were collected and weighed with electronic balance. The organs were flash-frozen with liquid nitrogen and stored at a temperature of −80 °C.

Automated Hematologic Analysis in Blood

In the present study, we used automatic hematology analyzer (Sysmex, Japan) to measure seven commonly-used erythroid-related hematologic parameters in EDTA-anticoagulated venous blood samples, including RBC count (× 1012/L), Hb concentration (g/L), hematocrit (HCT) (%), mean corpuscular volume (MCV) (fL), mean corpuscular hemoglobin concentration (MCHC) (g/L), RDW-coefficient of variation (RDW-CV) (%), and RDW-standard deviation (RDW-SD) (fL). These hematologic parameters were measured within 2 h after sample collection, and each blood sample was assayed in duplicate. The average levels for these erythroid-related hematologic parameters were included in the following statistical analyses.

For hematological analysis, citrated whole blood was analyzed using an automatic hematology analyzer (Sysmex, Kobe, Japan), following standard protocols. Red blood cells (RBCs), white blood cells (WBCs), platelets and hemoglobin were quantified automatically.

Diagnosis of Dengue Infection

Dengue NS1 antigen test (Panbio) was used to diagnose dengue. Detection of DENV RNA was performed with reverse transcription-PCR method as previously described.[10 (link)] RNA was extracted from 140 μl of plasma using QIAamp viral RNA mini kit (Qiagen, Germany) according to the manufacturer's instructions. Examination of blood parameters was performed using an automatic hematology analyzer (Sysmex, Japan). Blood electrolytes were determined using an automatic chemical analysis Cobas 501 (Roche, Germany).

Hematological Profiling of Aged Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood samples were collected from the mice when they were 95 weeks old (after 43 weeks of SMF exposure) using a blood lancet. Then, 200 μL of the blood samples were transferred to a tube with 0.15% (M/V) EDTA-K2-2H2O anticoagulant and subjected to a complete blood count by an automatic hematology analyzer (Sysmex, Japan). We measured the red blood cell, hematocrit, hemoglobin, platelet, white blood cell, neutrophil, eosinophil, monocyte, lymphocyte, mean corpuscular volume, mean platelet volume, plateletcrit, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, and platelet distribution width.

Hematology and Transaminase Analysis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Whole blood was obtained from the mice through their heart and collected in a potassium EDTA (Becton Dickinson, Franklin Lakes, New Jersey, USA) tube. Hematology profile was analyzed using an automatic hematology analyzer (Sysmex Corp., Japan). Transaminase enzymes alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were analyzed using the Lab kit from Chemelex, S. A. and the results were read at 340 nm wavelength on a spectrophotometer (Infinite 200 Pro, TECAN, Switzerland). The absorbance difference and the average absorbance difference were calculated and multiplied with 1750. Then, the concentration was expressed in units per liter.

Whole blood (K2EDTA tube) was collected from the wing vein before slaughter for routine hematological parameters using an automatic hematology analyzer (Sysmex, Japan), including white blood cells (WBCs), red blood cells (RBCs), hemoglobin (HGB), and platelets (PLTs). Serum was isolated from non-anticoagulant blood via centrifugation at 3,000 rpm/min for 10 min at 4°C for serum biochemical indicator determination. Serum total cholesterol (TC) and triglyceride (TG) levels were analyzed using a Fully Automatic Biochemical Analyzer (Hitachi, Japan).

The whole blood (K2EDTA-tube) was collected from the wing vein of NW, UAW, and BAW phenotype of HW geese (n = 9–10) for routine hematological parameter using the automatic hematology analyzer (Sysmex, Japan), including white blood cells (WBC), red blood cells (RBC), hemoglobin (HGB), neutrophils (NEUT), lymphocyte (LYMPH), monocytes (MONO), eosinophilic cell (EO), basophilic cell (BASO), hematocrit (HCT), mean corpuscular volume (MCV), mean corpuscular hemoglobin (MHC), mean corpuscular hemoglobin concentration (MCHC), platelet (PLT), thrombocytocrit (PCT), mean platelet volume (MPV), platelet ratio (P-LCR), and platelet distribution width (PDW). Serum was isolated from non-anticoagulant blood via centrifuged at 3,000 rpm/min for 10 min at 4°C for serum biochemical indicators determination. Serum total cholesterol (TC), high density lipoprotein (HDL), low density lipoprotein (LDL), triglycerides (TG), total protein (TP), albumin (ALB), globulin (GLB), Ca, P, creatine kinase (CK), alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) were analyzed using Fully Automatic Biochemical Analyzer (Hitachi, Japan).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
